【摘要】：目的:研究放療對(duì)人涎腺腺樣囊性癌細(xì)胞SACC-83和SACC-LM的影響以及與miRNA-21表達(dá)的相關(guān)性。方法:用Varian 23 EX對(duì)培養(yǎng)中的SACC-83和SACC-LM予從0(對(duì)照)、2、6、10 Gy照射,CCK-8檢測(cè)、流式細(xì)胞儀觀察放射處理后SACC-83、SACC-LM的細(xì)胞增殖和凋亡的變化;使用qRT-PCR法分析不同劑量放療后SACC-83、SACC-LM細(xì)胞miRNA-21的表達(dá)變化。結(jié)果:SACC-LM細(xì)胞放射處理48 h后增殖能力隨放療劑量的增大而降低(F=1321.646,P=0.000),早期凋亡率(F=354.484,P=0.039)、晚期凋亡率(F=254.278,P=0.042)和異倍體比例(F=1562.991,P=0.001)隨著放療劑量的增大而升高,且miRNA-21的相對(duì)表達(dá)量隨放療劑量的增大而減小(F=177.964,P=0.011);SACC-83細(xì)胞放療48 h后6 Gy放療組細(xì)胞增殖能力明顯低于其他各放療組(F=1537.214,P=0.013),6 Gy劑量下早期凋亡率(F=88.579,P=0.006)、晚期凋亡率(F=1391.345,P=0.033)、異倍體比例(F=250.461,P=0.004)最高,miRNA-21的相對(duì)表達(dá)量最低(F=134.868,P=0.017)。結(jié)論:miRNA-21可能與SACC-LM、SACC-83細(xì)胞的放療敏感性呈負(fù)相關(guān)。
[Abstract]:Aim: to study the effect of radiotherapy on SACC-83 and SACC-LM in human salivary adenoid cystic carcinoma cells and its correlation with miRNA-21 expression. Methods: SACC-83 and SACC-LM in culture were detected by Varian 23EX and the expression of miRNA-21 in SACC-83 SACC-LM cells was detected by flow cytometry after radiation treatment, and the expression of miRNA-21 in SACC-83 SACC-LM cells was analyzed by qRT-PCR method. Results after 48 hours of radiation treatment, the proliferative ability of the cells decreased with the increase of radiation dose, the early apoptosis rate was 354.484%, the late stage apoptosis rate was 254.278U (0.042) and the ratio of aneuploidy (F1562.991mP0.001) increased with the increase of radiation dose. And the relative expression of miRNA-21 decreased with the increase of radiation dose. The proliferative ability of SACC-83 cells in 6 Gy radiotherapy group was significantly lower than that in other radiotherapy groups at the dose of F1537.214P0.013 ~ 6 Gy, the late apoptosis rate was 1391.345P0. 033, and the ratio of aneuploidy was 0.033. The highest relative expression level of miRNA-21 was the lowest (FF134.868) P0. 017. Conclusion the results suggest that the sensitivity of SACC-LMN SACC-83 cells to radiotherapy is negatively correlated with the fraction miRNA-21.
