【摘要】:目的:以體外培養(yǎng)的人牙周膜成纖維細(xì)胞(hPDLFs)為研究對(duì)象,探討新型小分子RAGE特異性抑制劑對(duì)高糖環(huán)境培養(yǎng)下牙周膜成纖維細(xì)胞炎性因子的影響。方法:原代培養(yǎng)hPDLFs,Western-Blot檢測(cè)高糖對(duì)RAGE表達(dá)的影響,RT-PCR檢測(cè)FPS-ZM1對(duì)高糖環(huán)境下IL-6和TNF-α基因表達(dá)的影響,ELISA檢測(cè)IL-6和TNF-α的蛋白分泌變化。結(jié)果:高糖可導(dǎo)致hPDLFs表面RAGE表達(dá)增加,加重細(xì)胞炎癥反應(yīng)。而FPS-ZM1可以下調(diào)IL-6和TNF-α的基因表達(dá),同時(shí)減少炎癥介質(zhì)IL-6和TNF-α的分泌。結(jié)論:RAGE特異性抑制劑FPS-ZM1可以在一定程度上逆轉(zhuǎn)高糖刺激導(dǎo)致hPDLFs產(chǎn)生的炎癥反應(yīng)。
[Abstract]:Aim: to investigate the effect of a novel small molecule RAGE specific inhibitor on inflammatory cytokines of human periodontal ligament fibroblasts cultured in high glucose environment. Methods: the effect of high glucose on the expression of RAGE was detected by Western-Blot in primary culture. The effect of FPS-ZM1 on the expression of IL-6 and TNF- 偽 was detected by RT-PCR. The protein secretion of IL-6 and TNF- 偽 was detected by Elisa. Results: high glucose could increase the expression of RAGE on hPDLFs surface and aggravate the inflammatory reaction. FPS-ZM1 can down-regulate the gene expression of IL-6 and TNF- 偽, and decrease the secretion of IL-6 and TNF- 偽. Conclusion FPS-ZM1, a specific inhibitor of age, can reverse the inflammatory response of hPDLFs induced by high glucose stimulation to some extent.
