腺病毒介導HGF轉(zhuǎn)染脂肪干細胞復合溫敏型可注射水凝膠對兔顳下頜關節(jié)骨關節(jié)病髁突軟骨的修復作用

【摘要】:目的:探討肝細胞生長因子(HGF)轉(zhuǎn)染脂肪干細胞復合水凝膠支架材料對促進兔顳下頜關節(jié)炎病理狀態(tài)微環(huán)境中受損軟骨修復的意義。方法:培養(yǎng)兔脂肪干細胞,利用腺病毒介導對其進行HGF轉(zhuǎn)染并制備ADSCs/HGF/水凝膠支架材料。兔顳下頜關節(jié)上腔為注射區(qū)并分為5組,A組為空白對照組,B組為模型組,C組為單純細胞治療組,單純脂肪干細胞腔內(nèi)注射,D組為干細胞復合材料組,為造模后腔內(nèi)注射干細胞復合可注射水凝膠支架材料,E組為轉(zhuǎn)染后干細胞復合水凝膠注射。所有材料移植成功后各組動物分別于用藥第3、9周后取材,制備標本并行髁突軟骨掃描電鏡、組織學觀察及實時定量PCR檢測,采用SPSS17.0軟件包對數(shù)據(jù)進行方差分析及t檢驗。結果:掃描電鏡與組織學觀察可見,D、E組軟骨病變區(qū)改善情況優(yōu)于B、C組。與模型組比較,實時定量PCR結果顯示,E組MMP-13表達與空白組相對持平但低于C、D組,差異有顯著性(P0.05);E組TIMP-1表達與空白組相對持平但高于C、D組,差異有顯著性(P0.05)。結論:腺病毒介導HGF轉(zhuǎn)染脂肪干細胞復合可注射型水凝膠對兔顳下頜關節(jié)骨關節(jié)病受損軟骨具有明顯修復作用。
[Abstract]:Aim: to investigate the effect of HGF- transfected adipose stem cells (ASCs) combined with hydrogel scaffold on the repair of damaged cartilage in rabbit temporomandibular arthritis (TMJ) microenvironment.Methods: rabbit adipose stem cells were cultured and transfected with HGF mediated by adenovirus. ADSCs/HGF/ hydrogel scaffolds were prepared.Rabbit temporomandibular joint (TMJ) was divided into 5 groups: group A as blank control group, group B as model group, group C as simple cell therapy group, and group D as stem cell composite group after intracavitary injection of adipose stem cells.Group E was treated with transfection of stem cells combined with hydrogel injection.After all the materials were transplanted successfully, the animals were collected after the third week of treatment. The specimens were prepared with scanning electron microscope, histological observation and real-time quantitative PCR detection. The data were analyzed by SPSS17.0 software package and t-test.Results: scanning electron microscope and histological observation showed that the improvement of cartilage lesion in DU E group was better than that in BUC group.Compared with the model group, real-time quantitative PCR showed that the expression of MMP-13 in group E was relatively equal to that in the blank group but lower than that in group C D, and the difference was significant (P 0.05). The expression of TIMP-1 in group E was similar to that in group C (P 0.05), but higher than that in group C (D), and the difference was significant (P 0.05).Conclusion: Adenovirus-mediated HGF transfection of adipose stem cells combined with injectable hydrogel can repair the damaged cartilage of temporomandibular joint osteoarthritis in rabbits.
