【摘要】：目的探討脂多糖(LPS)對(duì)小鼠成骨細(xì)胞MC3T3-E1中基質(zhì)金屬蛋白酶-13(MMP-13)表達(dá)的影響及其分子機(jī)制。方法采用LPS處理傳代培養(yǎng)的小鼠成骨細(xì)胞MC3T3-E1,Western blotting檢測(cè)MC3T3-E1中MMP-13及轉(zhuǎn)錄因子C/EBPβ、核因子κB(NF-κB)及其抑制蛋白IκB、腫瘤壞死因子-α(TNF-α)、白介素-6(IL-6)的表達(dá);觀察使用NF-κB特異性抑制劑Bay11-7082干預(yù)及特異性siRNA沉默C/EBPβ、TNF-α和IL-6表達(dá)對(duì)MC3T3-E1中MMP-13表達(dá)的影響。結(jié)果 Western blotting檢測(cè)結(jié)果顯示:LPS能夠誘導(dǎo)MC3T3-E1中MMP-13高表達(dá),促進(jìn)NF-κB、C/EBPβ、TNF-α和IL-6的表達(dá)和活化,抑制IκB表達(dá);Bay11-7082干預(yù)及siRNA沉默TNF-α表達(dá)能有效抑制MC3T3-E1中TNF-α和MMP-13的表達(dá),而siRNA沉默C/EBPβ和IL-6表達(dá)對(duì)MC3T3-E1中MMP-13的表達(dá)無(wú)明顯影響。結(jié)論 LPS通過(guò)NF-κB/TNF-α誘導(dǎo)MMP-13在成骨細(xì)胞MC3T3-E1中高表達(dá),這為闡明牙周炎的發(fā)病機(jī)制奠定了基礎(chǔ)。
[Abstract]:Objective to investigate the effect of lipopolysaccharide (LPS) on the expression of matrix metalloproteinase-13 (MMP-13) in mouse osteoblasts (MC3T3-E1) and its molecular mechanism.Methods the expression of MMP-13, C/EBP 尾, NF- 魏 B, its inhibitor I 魏 B, tumor necrosis factor- 偽 -TNF- 偽, interleukin-6 (IL-6) in mouse osteoblasts treated with LPS were detected by Western blotting.To observe the effect of Bay11-7082 intervention and specific siRNA silencing the expression of C/EBP 尾 -TNF- 偽 and IL-6 on the expression of MMP-13 in MC3T3-E1.Results Western blotting assay showed that Western could induce high MMP-13 expression in MC3T3-E1, promote the expression and activation of NF- 魏 B C / EBP 尾 -TNF- 偽 and IL-6, inhibit the expression of I 魏 B and inhibit the expression of TNF- 偽 and MMP-13 in MC3T3-E1, and inhibit the expression of TNF- 偽 and MMP-13 in MC3T3-E1 by inhibiting the expression of TNF- 偽 by Bay11-7082 and silencing the expression of TNF- 偽 by siRNA.However, the silencing of C/EBP 尾 and IL-6 by siRNA had no significant effect on the expression of MMP-13 in MC3T3-E1.Conclusion LPS induces high expression of MMP-13 in osteoblast MC3T3-E1 through NF- 魏 B / TNF- 偽, which lays a foundation for elucidating the pathogenesis of periodontitis.
