DKK-1在顳下頜關(guān)節(jié)骨關(guān)節(jié)病中的作用

【摘要】:第一部分DKK-1蛋白在骨關(guān)節(jié)病滑膜中的表達(dá)目的:DKK-1在腫瘤、風(fēng)濕性關(guān)節(jié)炎等病理過(guò)程均發(fā)揮著重要作用,本部分研究目的在于探究DKK-1在顳下頜關(guān)節(jié)骨關(guān)節(jié)病滑膜中的表達(dá)。方法:利用免疫組織化學(xué)分別檢測(cè)DKK-1在顳下頜關(guān)節(jié)骨關(guān)節(jié)病滑膜中的表達(dá)水平;結(jié)果:多數(shù)顳下頜關(guān)節(jié)滑膜組織中均可檢測(cè)到DKK-1。正常組及骨關(guān)節(jié)病組的DKK-1的表達(dá)部位無(wú)明顯區(qū)別,均表現(xiàn)為胞質(zhì)和胞核的染色,并且在間質(zhì)區(qū)域亦有稍淺的染色。在染色強(qiáng)度上,骨關(guān)節(jié)病組的滑膜呈現(xiàn)出明顯的顏色加深。骨關(guān)節(jié)病組深棕色染色,細(xì)胞染色多而廣泛,而正;そM染色淺,部分細(xì)胞不染色。在大血管及新生小血管周圍DKK-1表達(dá)尤為加強(qiáng)。經(jīng)統(tǒng)計(jì)學(xué)分析,骨關(guān)節(jié)病組DKK-1表達(dá)明顯強(qiáng)于正;そM。結(jié)論:DKK-1蛋白在顳下頜關(guān)節(jié)骨關(guān)節(jié)病滑膜中高表達(dá)。第二部分DKK-1及VEGF蛋白在顳下頜關(guān)節(jié)紊亂病關(guān)節(jié)滑液中表達(dá)的表達(dá)目的:顳下頜關(guān)節(jié)滑液中蛋白含量的變化反應(yīng)了滑膜細(xì)胞病理、生理狀況。本部分研究目的在于檢測(cè)顳下頜關(guān)節(jié)滑液中DKK-1和VEGF的含量,并分析它們的相關(guān)性。方法:抽取正常志愿者、顳下頜關(guān)節(jié)盤移位、骨關(guān)節(jié)病患者的滑液,利用ELISA檢測(cè)顳下頜關(guān)節(jié)滑液中DKK-1及VEGF的含量。結(jié)果:DKK-1及VEGF蛋白在顳下頜關(guān)節(jié)病滑液中高表達(dá),在骨關(guān)節(jié)病組DKK-1、VEGF的表達(dá)水平最高,不可復(fù)性關(guān)節(jié)盤前移位次之,而可復(fù)性關(guān)節(jié)病組在患者人群中DKK-1表達(dá)最低,通過(guò)相關(guān)性分析,DKK-1與VEGF之間的表達(dá)呈正相關(guān)性結(jié)論:DKK-1及VEGF蛋白在顳下頜關(guān)節(jié)病關(guān)節(jié)滑液中高表達(dá)。DKK-1高表達(dá)可能與顳下頜關(guān)節(jié)病血管生成有關(guān)第三部分DKK-1在顳下頜關(guān)節(jié)滑膜細(xì)胞中的表達(dá)及作用機(jī)制目的:本部分研究目的在于通過(guò)顳下頜關(guān)節(jié)滑膜細(xì)胞原代培養(yǎng),在體外環(huán)境中檢測(cè)DKK-1對(duì)于VEGF表達(dá)的影響及其機(jī)制方法:對(duì)炎性環(huán)境下顳下頜關(guān)節(jié)滑膜細(xì)胞DKK-1表達(dá)進(jìn)行檢測(cè),并分析封閉DKK-1及給予外源性DKK-1后VEGF的表達(dá),采用HIF-1α siRNA干擾后檢測(cè)外源性DKK-1對(duì)VEGF表達(dá)的影響。利用條件培養(yǎng)基檢測(cè)DKK-1對(duì)血管內(nèi)皮細(xì)胞系遷移和成管能力的影響。結(jié)果:炎性環(huán)境中DKK-1及VEGF在顳下頜關(guān)節(jié)滑膜細(xì)胞中表達(dá)增加,顳下頜關(guān)節(jié)滑膜細(xì)胞中DKK-1通過(guò)上調(diào)HIF-1a來(lái)調(diào)控VEGF的表達(dá),DKK-1對(duì)臍靜脈內(nèi)皮細(xì)胞HUVECs遷移、成管能力具有上調(diào)的作用結(jié)論:DKK-1參與調(diào)控顳下頜關(guān)節(jié)滑膜細(xì)胞VEGF的表達(dá),并可能與滑膜炎癥血管生成有關(guān)。
[Abstract]:Part 1 expression of DKK-1 protein in synovial osteoarthritis Objective: DKK-1 in cancer, rheumatoid arthritis and other pathological processes play important roles, the purpose of this part of study is to explore DKK-1 expression in temporomandibular joint osteoarthritis synovium. Methods: the immunohistochemical expression of DKK-1 in temporomandibular joint osteoarthritis in the synovium were detected; results: most of the temporomandibular joint synovial tissue can be detected in the expression site of DKK-1. normal group and osteoarthropathy group DKK-1 showed no significant difference between staining of cytoplasm and nucleus, and in the interstitial region is also stained slightly lighter staining intensity in. On the synovial osteoarthritis group showed obvious color deepened. Bone and joint disease group deep brown staining, cell staining and more widely, and the normal synovial staining shallow, non staining. Some cells in large vessel and newborn Perivascular DKK-1 expression is particularly strengthened. By statistical analysis, bone and joint disease group DKK-1 expression was significantly stronger in normal synovial group. Conclusion: high expression of DKK-1 protein in temporomandibular joint synovial membrane. The second part of DKK-1 and VEGF protein expression in the objective table of temporomandibular disorders: changes of protein content in synovial fluid temporomandibular joint synovial fluid reaction in synovial cell pathological, physiological status. The purpose of this part of study is to detect the level of DKK-1 and VEGF in temporomandibular joint synovial fluid, and analyze their relationship. Methods: from normal volunteers, disc displacement of temporomandibular joint synovial fluid, bone joint disease patients, the contents of DKK-1 and VEGF ELISA detection temporomandibular joint synovial fluid. Results: DKK-1 and VEGF protein expression in temporomandibular joint disease in synovial fluid of osteoarthritis, in group DKK-1, the expression level of VEGF is the highest, the irreducible closed section Anterior disc displacement of temporomandibular joint disease and in patients of DKK-1 group was the lowest, through correlation analysis, the expression of positive correlation between DKK-1 and VEGF. Conclusion: DKK-1 and VEGF protein expression in temporomandibular joint disease in synovial fluid of high expression of.DKK-1 may be associated with temporomandibular joint disease objective function and expression of angiogenesis the third part of the DKK-1 mechanism in the temporomandibular joint synovial cells: the purpose of this part is through the study of temporomandibular joint synovial cells cultured in vitro environment for detection of DKK-1 VEGF expression and its mechanism methods: to detect the expression of inflammatory environment of temporomandibular joint synovial cell DKK-1, and analyze the expression of VEGF the closed DKK-1 and exogenous DKK-1, detect the effect of exogenous DKK-1 on the expression of VEGF by HIF-1 alpha after siRNA interference. Using the conditioned medium for detection of DKK-1 on blood vessels Effect of endothelial cell migration and tube formation. Results: DKK-1 and VEGF expression increased in the inflammatory environment in temporomandibular joint synovial cells in DKK-1 synovial cells by upregulation of HIF-1a in regulating the expression of VEGF, DKK-1, HUVECs on the migration of human umbilical vein endothelial cells, conclusion: tube has the ability to increase the expression of DKK-1 is involved in the regulation of temporomandibular joint synovial cells VEGF, and may be associated with synovitis of angiogenesis.
