【摘要】：目的:研究HOTAIR在舌鱗狀細(xì)胞癌中的表達(dá),并探究HOTAIR與舌鱗狀細(xì)胞癌淋巴結(jié)轉(zhuǎn)移的關(guān)系。方法:對(duì)19例舌鱗狀細(xì)胞癌患者的癌組織以及其對(duì)應(yīng)的19例癌旁組織進(jìn)行實(shí)時(shí)熒光定量PCR檢測(cè),檢測(cè)HOTAIR在癌組織、癌旁組織的相對(duì)表達(dá)量,把結(jié)果與患者的TNM分期進(jìn)行關(guān)聯(lián)比較,并進(jìn)一步通過(guò)構(gòu)建沉默HOTAIR表達(dá)的CAL27、SCC9細(xì)胞系,檢測(cè)腫瘤細(xì)胞侵襲、遷移能力的改變。結(jié)果:19對(duì)舌鱗狀細(xì)胞癌組織中,26.3%顯示出HOTAI的高表達(dá)(P=0.016 90.05),且高表達(dá)者均為具有淋巴結(jié)轉(zhuǎn)移的患者,而在無(wú)淋巴結(jié)轉(zhuǎn)移的癌組織中HOTAIR呈現(xiàn)低表達(dá)(P0.001),通過(guò)RNA干擾技術(shù)降低HOTAIR在CAL27及SCC9細(xì)胞系中的表達(dá)水平,結(jié)果發(fā)現(xiàn)細(xì)胞侵襲(P0.001)和遷移能力(P0.001)受到了抑制。結(jié)論:長(zhǎng)鏈非編碼RNA HOTAIR在舌鱗狀細(xì)胞癌淋巴結(jié)轉(zhuǎn)移過(guò)程中可能發(fā)揮促進(jìn)因子的作用。
[Abstract]:Objective: to study the expression of HOTAIR in squamous cell carcinoma of tongue. Methods: to investigate the relationship between HOTAIR and lymph node metastasis in tongue squamous cell carcinoma. Methods: 19 patients with tongue squamous cell carcinoma and 19 adjacent tissues were detected by real-time fluorescence quantitative PCR, and HOTAIR was detected in cancer tissue. The relative expression level of paracancerous tissues was correlated with the TNM stage of the patients. Furthermore, the invasion of tumor cells was detected by constructing a cell line CAL27, SCC9, which silenced the expression of HOTAIR. Results 26.3% of patients with squamous cell carcinoma of tongue showed high expression of HOTAI (0.016 90.05%), and the patients with high expression were all patients with lymph node metastasis. However, low expression of HOTAIR was found in cancer tissues without lymph node metastasis, and the expression of HOTAIR in CAL27 and SCC9 cell lines was decreased by RNA interference technique. The results showed that cell invasion and migration were inhibited. Conclusion: long chain noncoding RNA HOTAIR may play a role as a promoter in lymph node metastasis of tongue squamous cell carcinoma.
