【摘要】：目的:觀察煙草浸提液(ST)對(duì)人牙周膜細(xì)胞(hPDLCs)增殖、熱休克蛋白70(HSP70)表達(dá)的影響。方法:常規(guī)培養(yǎng)hPDLCs,第5~8代細(xì)胞用于實(shí)驗(yàn)。對(duì)照組為常規(guī)培養(yǎng)液,實(shí)驗(yàn)組為含不同濃度的ST培養(yǎng)液(0.016~50 g/L ST),作用24 h后MTT法檢測(cè)細(xì)胞增殖、免疫組化和Western Blot法檢測(cè)細(xì)胞HSP70的表達(dá)。結(jié)果:ST濃度為0.4~50 g/L時(shí)抑制hPDLCs增殖,增強(qiáng)細(xì)胞質(zhì)及胞核內(nèi)HSP70的表達(dá);ST濃度增加,其作用更明顯。結(jié)論:ST可抑制hPDLCs增殖,促進(jìn)HSP70呈表達(dá)。
[Abstract]:Objective: to observe the effect of tobacco extract (STS) on the proliferation and expression of heat shock protein 70 (HSP70) in human periodontal ligament cells. The experimental group was treated with different concentrations of ST-containing 0.016 ~ 50 g / L STN. The proliferation of hPDLCs was detected by MTT method after 24 h exposure, and the expression of HSP70 was detected by immunohistochemistry and Western Blot method. Results the proliferation of hPDLCs was inhibited when the concentration of St was 0.4g / L, and the expression of HSP70 was detected by Western Blot method. The increased expression of HSP70 in cytoplasm and nucleus increased the concentration of St, and the effect was more obvious. Conclusion: 1. Conclusion: 1. St can inhibit the proliferation of hPDLCs and promote the expression of HSP70.
