響應(yīng)面法優(yōu)化IgY抑制具核梭桿菌增殖的研究

【摘要】:目的:利用響應(yīng)面法研究免疫球蛋白Y(IgY)影響口腔唾液模擬環(huán)境中具核梭桿菌(F.nucleatum)的生長繁殖特性,發(fā)現(xiàn)F.nucleatum最終菌落數(shù)與培養(yǎng)時間、F.nucleatum初始接種濃度和IgY劑量的關(guān)系。方法:以F.nucleatum最終菌落數(shù)為評價指標(biāo),在單因素試驗基礎(chǔ)上,設(shè)計三因素三水平響應(yīng)面實驗進(jìn)行優(yōu)化,通過響應(yīng)面法對回歸方程的分析,得到各因素最佳條件,優(yōu)化后的條件重復(fù)試驗3次,以驗證模型的準(zhǔn)確性。結(jié)果:以2×10~6CFU/mL初始濃度接種F.nucleatum、181mmol/L劑量給藥IgY,培養(yǎng)時間6h得到最終菌落數(shù)4.52×10~4 CFU/mL,與預(yù)測值相對誤差小于5%,方差分析表明模型P值顯著,缺失值P值不顯著,理論值與實測值擬合度良好,優(yōu)化后F.nucleatum最終菌落數(shù)顯著降低。結(jié)論:響應(yīng)面法優(yōu)化結(jié)果表明IgY能顯著抑制口腔唾液模擬環(huán)境中具核梭桿菌的增殖,回歸模型能夠預(yù)測IgY影響口腔唾液模擬環(huán)境中F.nucleatum增殖的特性。
[Abstract]:Objective: to study the growth and reproduction characteristics of F. nucleatum (F. nucleatum) in oral saliva simulated environment affected by immunoglobulin YG IgY by using response surface method (RSM). The relationship between the final colony number of F. nucleatum and the initial inoculation concentration of F. nucleatum and the dose of IgY was found. Methods: based on the single factor test, three factors and three levels of response surface test were designed to optimize F. nucleatum final colony number. Through the analysis of the regression equation by the response surface method, the optimum conditions of each factor are obtained, and the optimized conditions are repeated for 3 times. Results: at the initial concentration of 2 脳 10 ~ (-6) CFU / mL, the final colony number was 4.52 脳 10 ~ (4) CFU / mL, and the relative error from the predicted value was less than 5. The variance analysis showed that the model P value was significant, the missing value P value was not significant, and the final colony number was 4.52 脳 10 ~ (4) CFU / mL for 6 h. Conclusion: the response surface method showed that IgY could significantly inhibit the proliferation of Clostridium nucleatum in oral saliva simulated environment. [WT5 "HZ] [WT5" BZ] [WT5 "BZ] [WT5" BZ] [WT5BZ] the results showed that IgY could significantly inhibit the proliferation of Clostridium nucleatum. The regression model can predict the effect of IgY on the proliferation of F. nucleatum in oral saliva simulation environment.
