紅景天苷對成骨細胞的作用及相關分子機制研究

【摘要】:目的:研究紅景天苷(SAL)對人成骨樣MG-63細胞增殖、分化和HIF-1α依賴的VEGF表達的影響及相關分子機制。方法:采用MTS法、流式細胞術分析、RT-PCR、ELISA、Western blot等技術觀察SAL對MG-63細胞增殖、分化和HIF-1α依賴的VEGF表達的影響,進一步探討其相關分子機制。結果:SAL通過調控MG-63細胞的周期分布及上調Runx2和Osterix表達促進成骨細胞的增殖和分化。另外,SAL可明顯上調下游靶基因VEGF的表達。結論:SAL可顯著促進成骨樣MG-63細胞的增殖、分化以及HIF-1α依賴的VEGF的表達,進而促進骨和血管新生。
[Abstract]:Objective: to study the effect of salidroside on the proliferation, differentiation and HIF-1 偽 dependent VEGF expression of human osteoblast like MG-63 cells and its molecular mechanism. Methods: the proliferation of MG-63 cells was observed by MTS and flow cytometry. Differentiation and the effect of HIF-1 偽 -dependent VEGF expression, Results the cell cycle distribution of MG-63 cells was regulated and the expression of Runx2 and Osterix was upregulated to promote the proliferation and differentiation of osteoblasts. In addition, Sal could significantly up-regulate the expression of downstream target gene VEGF. Conclusion\% Sal can promote the proliferation and differentiation of osteoblasts by regulating the cell cycle distribution and upregulating the expression of Runx2 and Osterix. It can significantly promote the proliferation of osteoblast-like MG-63 cells. Differentiation and HIF-1 偽-dependent VEGF expression promote bone and angiogenesis.
