高濃度葡萄糖對人牙周膜成纖維細(xì)胞成骨分化能力的影響

【摘要】:目的研究高葡萄糖濃度刺激下牙周膜成纖維細(xì)胞(PDLC)的增殖能力和成骨分化能力。方法體外組織塊法培養(yǎng)非糖尿病患者PDLC,分別用0 mg·L-1(C組)、1 100 mg·L-1(L組)、4 500 mg·L-1(H組)濃度葡萄糖刺激PDLC,噻唑藍(MTT)法檢測PDLC增殖能力,礦化誘導(dǎo)后茜素紅染色觀察礦化結(jié)節(jié)的形成情況,堿性磷酸酶(ALP)活性檢測和實時定量聚合酶鏈反應(yīng)(RT-PCR)檢測成骨相關(guān)基因表達。結(jié)果 MTT檢測結(jié)果顯示,H組OD值較L組和C組低(P0.05);成骨誘導(dǎo)21 d后,H組礦化結(jié)節(jié)面積較L組和C組少(P0.05);成骨誘導(dǎo)期間,H組ALP活性較L組和C組明顯偏低(P0.05);H組早期成骨基因Runt相關(guān)轉(zhuǎn)錄因子2、ALP和Ⅰ型膠原誘導(dǎo)前后相對倍增數(shù)較L組和C組低(P0.05)。結(jié)論高濃度葡萄糖能夠抑制PDLC增殖能力和成骨分化能力。
[Abstract]:Objective to study the proliferation and osteogenic differentiation of periodontal ligament fibroblasts (PDLC) stimulated by high glucose concentration. PDLC was stimulated by glucose in 0 mg 路L ~ (-1) C group (n = 1 100 mg 路L ~ (-1)) L group (n = 4 500 mg 路L ~ (-1)). The proliferative ability of PDLC was detected by thiazolyl blue method, and the formation of mineralized nodules was observed by alizarin red staining after mineralization induction. Alkaline phosphatase (ALP) activity and real-time quantitative polymerase chain reaction (RT-PCR) were used to detect the expression of osteoblast-associated genes. The OD value of group H was lower than that of group L and group C (P 0.05). 21 days after osteogenesis induction, the area of mineralized nodules in group H was less than that in group L and group C (P 0.05). During osteogenic induction, the activity of ALP in group H was significantly lower than that in group L and group C (P 0.05). Group H had early osteogenic gene Runt related transcription factor 2. The relative doubling number of ALP and collagen I before and after induction was lower than that of group L and group C. ConclusionHigh concentration of glucose can inhibit the proliferation and osteogenic differentiation of PDLC.
