【摘要】:目的:檢測P-gp與HIF-1α在口腔鱗狀細(xì)胞癌癌組織中的表達(dá)及相互關(guān)系,為臨床評(píng)估口腔鱗癌化療耐藥性提供參考。方法:1、逆轉(zhuǎn)錄—多聚酶鏈反應(yīng)(RT-PCR)檢測口腔鱗癌癌組織P-gp(mdrl)基因及HIF-1 α基因在其中的表達(dá)。2、免疫組織化學(xué)SP法檢測口腔鱗癌癌組織HIF-1α蛋白和P-gp蛋白在其中的表達(dá)。結(jié)果:1、28例OSCC癌標(biāo)本,經(jīng)測定符合實(shí)驗(yàn)要求的有15例。電泳結(jié)果:P-gp(mdrl)基因表達(dá)為15例,HIF-1α表達(dá)為13例。兩者共表達(dá)有13例。列聯(lián)相關(guān)性分析經(jīng)卡方檢驗(yàn)確切概率法得出結(jié)果,有統(tǒng)計(jì)學(xué)意義(P0.05)。兩種基因在OSCC癌組織中的表達(dá)有正相關(guān)性,r=0.867。2、28例OSCC癌組織中HIF-1α蛋白陽性表達(dá)有21例(75%),P-gp蛋白陽性表達(dá)有20例(71.4%)。HIF-1α和P-gp表達(dá)在患者性別、腫瘤TNM分期、病理分級(jí)及淋巴結(jié)轉(zhuǎn)移比較統(tǒng)計(jì)學(xué)無意義(P0.05)。HIF-1α蛋白表達(dá)與腫瘤大小(P=0.029)、年齡(P=0.041)存在差別,P-gp蛋白表達(dá)與腫瘤大小(P=0.007)存在差別,統(tǒng)計(jì)學(xué)有意義(P0.05)。28例OSCC組織中兩者有陽性表達(dá)一致性共18例,列聯(lián)相關(guān)性分析經(jīng)卡方檢驗(yàn)確切概率法得出結(jié)果,兩者之間有統(tǒng)計(jì)學(xué)意義(P0.05),兩者在OSCC癌組織中的表達(dá)有正相關(guān)性,r=0.548。結(jié)論:P-gp和HIF-1α在OSCC癌組織中同時(shí)表達(dá)增高,兩者具有顯著相關(guān)性,可能協(xié)同介導(dǎo)OSCC原發(fā)性耐藥,其機(jī)制可能是通過HIF-1α上調(diào)P-gp表達(dá)來實(shí)現(xiàn)。P-gp和HIF-1α表達(dá)檢測可為臨床OSCC患者化療耐藥性評(píng)估提供參考指標(biāo)。
[Abstract]:Objective: to detect the expression of P-gp and HIF-1 偽 in oral squamous cell carcinoma (OSCC) and their relationship, and to provide a reference for clinical evaluation of chemotherapeutic resistance in oral squamous cell carcinoma (OSCC). Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of P-gpmdrl) gene and HIF-1 偽 gene in oral squamous cell carcinoma (OSCC). Immunohistochemical SP method was used to detect the expression of HIF-1 偽 protein and P-gp protein in oral squamous cell carcinoma (OSCC). 15 cases met the requirements of the experiment and 15 cases were detected by electrophoretic analysis. The expression of HIF-1 偽 was found in 13 cases, and 13 cases were co-expressed. The results were obtained by the chi-square test and the exact probability method. There was a positive correlation between the expression of the two genes in OSCC tissues. There were 21 cases of positive expression of HIF-1 偽 protein in 28 cases of OSCC carcinoma. The positive expression of P-gp protein in 20 cases was 71.4%. HIF-1 偽 and P-gp were expressed in sex and TNM stage of tumor. There was no significant difference in pathological grade and lymph node metastasis between the expression of P0.05U. HIF-1 偽 protein and the size of the tumor (P0. 029, age: P0. 041). There was a significant difference between the expression of P-gp protein and tumor size (P0. 007). The positive expression of P-gp protein was consistent in 18 cases in 28 cases of OSCC. The results obtained by the exact probability method of chi-square test showed that there was a significant difference between the two methods and there was a positive correlation between them in OSCC cancer tissues. Conclusion the expression of HIF-1 偽 and P-gp in OSCC cancer tissues is increased simultaneously, which has a significant correlation, and may co-mediate the primary drug resistance of OSCC. The mechanism may be that the expression of. P-gp and HIF-1 偽 can be detected by up-regulating the expression of P-gp by HIF-1 偽, which may provide a reference index for the evaluation of chemoresistance in patients with OSCC.
