【摘要】:精神分裂癥是一種異質(zhì)性精神障礙,目前多數(shù)學(xué)者認(rèn)為該病由遺傳及環(huán)境風(fēng)險(xiǎn)因子共同致病.傳統(tǒng)的遺傳學(xué)研究識(shí)別了一些精神分裂癥的候選基因,然而,外顯率及比值比較低,以及可重復(fù)性較差,使得這些候選基因在精神分裂癥發(fā)病機(jī)制中的角色受到限制.精神分裂癥的流行病學(xué)研究發(fā)現(xiàn)了一些可能與該病相關(guān)的環(huán)境風(fēng)險(xiǎn)因子,但仍受到方法學(xué)的限制以及互相矛盾的流行病學(xué)調(diào)查結(jié)果的困擾.目前,在環(huán)境與基因中間起調(diào)解作用的表觀(guān)遺傳學(xué),可能在精神分裂癥的發(fā)病機(jī)制中起重要作用.DNA甲基化是最穩(wěn)定且研究最深入的一種表觀(guān)遺傳學(xué)修飾.本文簡(jiǎn)要介紹DNA甲基化機(jī)制,主要評(píng)述精神分裂癥中基因組及特異位點(diǎn)如候選基因Reelin與COMT甲基化研究現(xiàn)狀,以及DNA甲基化在精神分裂癥中的研究困境.
