基于加權(quán)基因共表達(dá)網(wǎng)絡(luò)分析阿爾茨海默病相關(guān)核心靶點(diǎn)(英文)

目的阿爾茨海默。ˋlzheimer‘s disease,AD)是癡呆癥最常見的病因,但其發(fā)病機(jī)制尚不明確。本研究擬通過加權(quán)基因共表達(dá)網(wǎng)絡(luò)探究AD發(fā)病相關(guān)的核心靶點(diǎn)。方法 GSE36980數(shù)據(jù)集下載于高通量基因表達(dá)數(shù)據(jù)庫(Gene Expression Omnibus, GEO)。首先進(jìn)行數(shù)據(jù)標(biāo)準(zhǔn)化,質(zhì)控和過濾,以及計(jì)算軟閾值,然后根據(jù)基因表達(dá)的相關(guān)性,聚類劃分為不同的模塊,通過計(jì)算各模塊與臨床特征的相關(guān)系數(shù),確定關(guān)鍵基因模塊。我們通過基因富集分析(Gene Ontology,GO)和通路富集分析(Kyoto Encyclopedia of Genes and Genomes, KEGG)探究關(guān)鍵基因模塊中的基因功能。隨后通過STRING數(shù)據(jù)庫構(gòu)建蛋白-蛋白相互作用網(wǎng)絡(luò),并使用Cytoscape軟件MCODE插件進(jìn)行網(wǎng)絡(luò)拓?fù)浞治龊Y選核心調(diào)控基因,最后使用GEO外部數(shù)據(jù)集GSE1297和GSE28146對(duì)核心基因進(jìn)行驗(yàn)證。結(jié)果共表達(dá)基因共聚類27個(gè)模塊,其中6個(gè)模塊與AD發(fā)病顯著相關(guān),以此作為關(guān)鍵模塊用于下游分析。通過基因功能富集分析發(fā)現(xiàn)關(guān)鍵模塊與神經(jīng)遞質(zhì)傳遞(GO:0007268)、三...
