基于加權基因共表達網(wǎng)絡分析阿爾茨海默病相關核心靶點(英文)

目的阿爾茨海默病(Alzheimer‘s disease,AD)是癡呆癥最常見的病因,但其發(fā)病機制尚不明確。本研究擬通過加權基因共表達網(wǎng)絡探究AD發(fā)病相關的核心靶點。方法 GSE36980數(shù)據(jù)集下載于高通量基因表達數(shù)據(jù)庫(Gene Expression Omnibus, GEO)。首先進行數(shù)據(jù)標準化,質控和過濾,以及計算軟閾值,然后根據(jù)基因表達的相關性,聚類劃分為不同的模塊,通過計算各模塊與臨床特征的相關系數(shù),確定關鍵基因模塊。我們通過基因富集分析(Gene Ontology,GO)和通路富集分析(Kyoto Encyclopedia of Genes and Genomes, KEGG)探究關鍵基因模塊中的基因功能。隨后通過STRING數(shù)據(jù)庫構建蛋白-蛋白相互作用網(wǎng)絡,并使用Cytoscape軟件MCODE插件進行網(wǎng)絡拓撲分析篩選核心調控基因,最后使用GEO外部數(shù)據(jù)集GSE1297和GSE28146對核心基因進行驗證。結果共表達基因共聚類27個模塊,其中6個模塊與AD發(fā)病顯著相關,以此作為關鍵模塊用于下游分析。通過基因功能富集分析發(fā)現(xiàn)關鍵模塊與神經(jīng)遞質傳遞(GO:0007268)、三...
