CRH-CRHR1/GABA信號(hào)通路與抑郁癥發(fā)病的分子機(jī)制

抑郁癥是精神疾病中的主要難題之一。近年來由應(yīng)激引發(fā)的神經(jīng)內(nèi)分泌系統(tǒng)的改變在抑郁癥發(fā)病過程中的作用倍受關(guān)注,其中最重要的病理改變是下丘腦-垂體-腎上腺(Hypothalamic-Pituitary-Adrenocotical, HPA)軸的過度驅(qū)動(dòng)與功能亢進(jìn)(Hyper-activation)。HPA軸激活反應(yīng)的核心驅(qū)動(dòng)力來自位于下丘腦室旁核(Paraventricular nucleus, PVN)部位的促腎上腺皮質(zhì)激素釋放激素(Corticotropin Releasing Hormone, CRH,在嚙齒類中,稱作Corticotropin-Releasing Factor, CRF)神經(jīng)元釋放的CRH,再通過與其受體結(jié)合并激活下游級聯(lián)信號(hào)通路,引發(fā)HPA軸中各個(gè)功效環(huán)節(jié)應(yīng)激激素的大量釋放,在抑郁癥的發(fā)病中起著關(guān)鍵性作用。臨床結(jié)果顯示,對抑郁癥患者和健康人群給予抗抑郁藥物治療后,CRH基因的表達(dá)量與HPA軸的活性都相應(yīng)降低,提示抗抑郁藥療效可能與CRH及其Ⅰ型受體(coticotropin-releasing hormone receptor 1, CRHR1)相關(guān)。本研究主要從P...
