【摘要】：目的研究丙戊酸鈉(VPA)對(duì)岡田酸(OA)處理的人神經(jīng)母細(xì)胞瘤系(SH-SY5Y)細(xì)胞中tau蛋白磷酸化的影響。方法 MTT觀察OA對(duì)SH-SY5Y細(xì)胞活力的影響,制備tau蛋白過(guò)度磷酸化的細(xì)胞模型;Western blot檢測(cè)OA及VPA對(duì)Thr231位點(diǎn)tau蛋白磷酸化的影響。結(jié)果 MTT結(jié)果顯示,當(dāng)OA濃度大于40 nmol/L時(shí),細(xì)胞活力受到明顯抑制,而低于此濃度的OA對(duì)細(xì)胞活力的影響不明顯;Western blotting結(jié)果顯示,SH-SY5Y細(xì)胞經(jīng)OA(40 nmol/L,12 h)處理后,Tau蛋白磷酸化的水平明顯增加,給予VPA(10 mmol/L)作用12 h后,Tau蛋白磷酸化的水平明顯下降。結(jié)論 VPA可以抑制OA處理的SH-SY5Y細(xì)胞的tau蛋白過(guò)度磷酸化。
[Abstract]:Objective to study the effect of sodium valproate (VPA) on phosphorylation of tau protein in human neuroblastoma cell line (SH-SY5Y) treated with (OA). Methods the effect of OA on the activity of SH-SY5Y cells was observed by MTT. The cell model of tau protein hyperphosphorylation was prepared to detect the effect of OA and VPA on tau protein phosphorylation at Thr231 site. Results the results of MTT showed that when the concentration of OA was more than 40 nmol/L, the cell activity was significantly inhibited, but the effect of OA below this concentration on cell viability was not obvious.; Western blotting results showed that the phosphorylation level of Tau protein in SH-SY5Y cells treated with OA (40 nmol/L,12 h) increased significantly, and the phosphorylation level of Tau protein decreased significantly after treatment with VPA (10 mmol/L) for 12 h. Conclusion VPA can inhibit the hyperphosphorylation of tau protein in SH-SY5Y cells treated with OA.
