阿爾茨海默病治療性單鏈抗體的研究進(jìn)展

【摘要】:阿爾茨海默氏病(AD)的免疫治療方法自提出以來盡管經(jīng)歷了些許挫折,但經(jīng)過研究人員多年不懈努力,已經(jīng)得到了快速發(fā)展。其中針對β-淀粉樣蛋白的單鏈抗體(scFv)已經(jīng)成為AD治療性抗體中最有希望實(shí)現(xiàn)突破的一種策略,并正在成為免疫治療AD的抗體藥物研究的前沿和熱點(diǎn)。本文概述了AD治療性scFv的來源優(yōu)勢,對AD致病抗原識別與結(jié)合的特點(diǎn),改善AD病理特征的效應(yīng)以及與其效應(yīng)相關(guān)的腦轉(zhuǎn)運(yùn)機(jī)制和體外親和力成熟過程和結(jié)構(gòu)因素,旨在為相關(guān)研究提供理論線索和發(fā)展思路。
[Abstract]:Although the immunotherapy for Alzheimer's disease (AD) has experienced some setbacks since it was put forward, it has been developed rapidly after years of unremitting efforts by researchers. Single chain antibody against 尾-amyloid protein (scFv) has become one of the most promising strategies to achieve breakthrough in therapeutic antibody of AD, and is becoming the frontier and hot spot in the research of antibody drugs in immunotherapy of AD. In this paper, the source advantages of AD therapeutic scFv, the characteristics of recognition and combination of AD pathogenic antigens, the effect of improving the pathological features of AD, the mechanism of brain transport related to the effect, and the process and structural factors of in vitro affinity maturation were reviewed. The purpose of this paper is to provide theoretical clues and development ideas for related research.
