【摘要】：NR3C1參與HPA軸調(diào)節(jié),與應(yīng)激相關(guān)的身心疾病關(guān)系密切。以往針對(duì)NR3C1與焦慮障礙的研究基本以成人為考察對(duì)象,且大多關(guān)注負(fù)性生活事件等個(gè)體層面環(huán)境變量。本研究采用病例-對(duì)照設(shè)計(jì),以238名青少年為被試(焦慮障礙117人,對(duì)照121人),旨在考察NR3C1常見(jiàn)位點(diǎn)rs6191、rs6196、rs41423247的多態(tài)性、單倍型以及父母不同類型教養(yǎng)方式對(duì)焦慮障礙的影響。結(jié)果表明rs6191 GG基因型、rs6196 AA基因型、rs41423247 GG基因型與低焦慮障礙風(fēng)險(xiǎn)相關(guān)。父母過(guò)多的過(guò)度保護(hù)與冷漠拒絕、父親過(guò)少的溫暖關(guān)懷可預(yù)測(cè)較高的焦慮障礙風(fēng)險(xiǎn)。rs41423247多態(tài)性與母親溫暖關(guān)懷存在交互作用:rs41423247 GG基因型只在母親溫暖關(guān)懷較多時(shí)能降低焦慮障礙風(fēng)險(xiǎn),在母親溫暖關(guān)懷較少時(shí)與焦慮障礙并無(wú)顯著相關(guān)。此外,rs6191-rs6196-rs41423247構(gòu)成的單倍型GAG與低焦慮障礙風(fēng)險(xiǎn)顯著關(guān)聯(lián),TGC與高焦慮障礙風(fēng)險(xiǎn)顯著關(guān)聯(lián),且二者與母親溫暖關(guān)懷、母親過(guò)度保護(hù)分別存在交互作用。未來(lái)可采用追蹤研究,從表觀遺傳層面探討教養(yǎng)方式與NR3C1影響焦慮障礙的內(nèi)在機(jī)制。
[Abstract]:NR3C1 participates in the regulation of HPA axis and is closely related to stress-related physical and mental diseases. Previous studies on NR3C1 and anxiety disorders have focused on adults and most of them focused on negative life events and other individual environmental variables. A case-control study was conducted in 238 adolescents (117 with anxiety disorder and 121 as control) to investigate the polymorphism of rs6191,rs6196,rs41423247 at common loci of NR3C1. The effects of haplotypes and different parenting styles on anxiety disorder. The results showed that rs6191 GG genotype, rs6196 AA genotype and rs41423247 GG genotype were associated with low anxiety disorder risk. Too much protection and indifference from parents, The higher risk of anxiety disorder could be predicted by too little warm care of father. There was interaction between rs41423247 polymorphism and warm care of mother: rs41423247 GG genotype could reduce the risk of anxiety disorder only when the mother had more warm care. There was no significant correlation between anxiety disorder and maternal warmth. In addition, the haplotype GAG composed of rs6191-rs6196-rs41423247 was significantly associated with the risk of low anxiety disorder, and TGC was significantly associated with the risk of high anxiety disorder, and there was interaction between the haplotype GAG and the mother's warm care and mother's overprotection, respectively. In the future, tracing research can be used to explore the relationship between upbringing style and the internal mechanism of NR3C1 affecting anxiety disorder from the epigenetic level.
