創(chuàng)傷后應(yīng)激障礙大鼠海馬、杏仁核神經(jīng)元自噬和凋亡改變!

【摘要】:目的觀察創(chuàng)傷后應(yīng)激障礙(PTSD)模型大鼠海馬、杏仁核神經(jīng)元自噬和凋亡的改變,探討PTSD海馬、杏仁核體積異常的機(jī)制。方法成年健康雄性SD大鼠40只,隨機(jī)分為對(duì)照組和模型組。采用改良的單一連續(xù)應(yīng)激方法制備PTSD大鼠模型;采用Western blotting方法檢測(cè)海馬、杏仁核組織LC-3和Beclin-1的表達(dá)水平;TUNEL染色和流式細(xì)胞術(shù)檢測(cè)海馬、杏仁核神經(jīng)元凋亡情況。結(jié)果模型組海馬、杏仁核組織LC3-Ⅱ/LC3-Ⅰ比值和Beclin-1表達(dá)水平高于對(duì)照組(P0.05);模型組海馬、杏仁核神經(jīng)元TUNEL陽(yáng)性細(xì)胞百分率和凋亡率高于對(duì)照組(P0.01)。結(jié)論 PTSD模型大鼠海馬、杏仁核神經(jīng)元存在明顯的細(xì)胞自噬和凋亡,這可能是海馬、杏仁核體積異常的機(jī)制之一。
[Abstract]:Objective to observe the changes of autophagy and apoptosis of hippocampal and amygdaloid neurons in rats with post-traumatic stress disorder (PTSD) and to explore the mechanism of abnormal volume of hippocampus and amygdala in PTSD.
