糖皮質(zhì)激素受體:抑郁癥治療的潛在靶標(biāo)

【摘要】:抑郁癥是以顯著而持久的心境低落為主要特征的情感性精神障礙,其病理生理學(xué)機(jī)制復(fù)雜,涉及一系列神經(jīng)內(nèi)分泌的改變。糖皮質(zhì)激素(glucocorticoid,GC)水平升高、下丘腦-垂體-腎上腺軸(hypothalamic-pituitaryadrenal axis,HPA軸)功能亢進(jìn)是抑郁癥典型的病理生理改變之一,其中糖皮質(zhì)激素受體(glucocorticoid receptor,GR)在這一過程中起著關(guān)鍵性調(diào)節(jié)作用。增加GR的表達(dá)與功能,從而恢復(fù)HPA軸負(fù)反饋功能是抗抑郁劑發(fā)揮效應(yīng)的重要機(jī)制之一。該文綜述了近年來GR與抑郁癥的相關(guān)研究進(jìn)展,冀望為抗抑郁藥物研制提供新的思路。
[Abstract]:Depression is a kind of affective mental disorder characterized by significant and persistent depression. Its pathophysiological mechanism is complex and involves a series of neuroendocrine changes. The elevation of glucocorticoid (glucocorticoid,GC) and the hyperfunction of hypothalamus-pituitary-adrenal axis (hypothalamic-pituitaryadrenal axis,HPA axis) are typical pathophysiological changes of depression. The glucocorticoid receptor (glucocorticoid receptor, is one of the typical pathophysiological changes in depression. GR) plays a key role in this process. Increasing the expression and function of GR and restoring the negative feedback function of HPA axis is one of the important mechanisms of antidepressant effect. This paper reviews the recent progress in the research of GR and depression, hoping to provide new ideas for the development of antidepressants.
