GABRB2基因啟動(dòng)子區(qū)DNA甲基化與精神分裂癥及其治療的關(guān)系

【摘要】:目的探討γ-氨基丁酸A型受體β2亞基(gamma-aminobutyric acid type A receptor beta2 subunit,GABRB2)基因啟動(dòng)子區(qū)DNA甲基化水平與精神分裂癥及其藥物治療的關(guān)系。方法納入精神分裂癥患者53例和正常對(duì)照53名,采用Mass ARRY Epi TYPER DNA甲基化分析技術(shù)檢測(cè)所有受試者外周血GABRB2基因啟動(dòng)子區(qū)DNA甲基化水平。患者組中12例在藥物治療8周后再次檢測(cè)該基因啟動(dòng)子區(qū)DNA甲基化水平。結(jié)果患者組與對(duì)照組外周血GABRB2基因啟動(dòng)子區(qū)總甲基化和各Cp G位點(diǎn)的甲基化水平組間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。經(jīng)性別分層分析,患者組男性Cp G_28位點(diǎn)的甲基化水平較對(duì)照組男性降低[(0.215±0.084)vs.(0.264±0.103),P0.05]。治療后患者組外周血GABRB2基因啟動(dòng)子區(qū)總DNA甲基化水平較治療前降低[(0.088±0.037)vs.(0.121±0.063),P0.01],治療前后各Cp G位點(diǎn)甲基化水平差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論抗精神病藥物治療可降低精神分裂癥患者外周血中GABRB2基因啟動(dòng)子區(qū)DNA甲基化水平,提示基因啟動(dòng)子區(qū)DNA甲基化可能參與抗精神病藥物治療的作用機(jī)制。
[Abstract]:Objective to investigate the relationship between the level of DNA methylation in promoter region of 緯 -aminobutyric acid type A receptor 尾 2 subunit (gamma-aminobutyric acid type A receptor beta2 subunit,GABRB2) gene and schizophrenia and its drug therapy. Methods the DNA methylation levels in the promoter region of GABRB2 gene in peripheral blood of 53 patients with schizophrenia and 53 normal controls were detected by Mass ARRY Epi TYPER DNA methylation analysis. DNA methylation in the promoter region of the gene was detected again in 12 patients after 8 weeks of drug therapy. Results there was no significant difference in the total methylation of the promoter region of GABRB2 gene and the methylation level of each Cp G locus between the patients and the control group (P0.05). By sex stratification analysis, the methylation level of Cp G28 locus in the patient group was lower than that in the control group [(0.215 鹵0.084) vs. (0.264 鹵0.103), P0.05]. After treatment, the total DNA methylation level in the promoter region of GABRB2 gene in peripheral blood of the patients was lower than that before treatment [(0.088 鹵0.037) vs. (0.121 鹵0.063), P0.01]. There was no significant difference in the methylation level of each Cp G locus before and after treatment (P0.05). Conclusion antipsychotic therapy can reduce the level of DNA methylation in the promoter region of GABRB2 gene in the peripheral blood of patients with schizophrenia, suggesting that DNA methylation in the promoter region may be involved in the mechanism of antipsychotic drug therapy.
