【摘要】：目的研究阿爾茨海默病(AD)患者血小板脂質(zhì)筏內(nèi)神經(jīng)節(jié)苷脂GM1含量的變化,尋求一種可能作為輔助診斷AD的早期生物學(xué)指標(biāo)。方法選擇42例AD患者和45例認(rèn)知功能正常的健康對(duì)照者分別進(jìn)行神經(jīng)功能評(píng)定,采用以O(shè)ptiprep為介質(zhì)的密度梯度離心方法分離血小板脂質(zhì)筏,應(yīng)用點(diǎn)印跡的方法進(jìn)行鑒定后,采用BCA法測(cè)定血小板脂質(zhì)筏內(nèi)蛋白含量,比色法及點(diǎn)印跡定量分析法測(cè)定其內(nèi)神經(jīng)節(jié)苷脂GM1含量。結(jié)果在離心管25%~30%Optiprep溶液界面處可見(jiàn)一條白色的閃亮環(huán)帶,經(jīng)點(diǎn)印跡法證實(shí),位于第6層的此白色閃亮環(huán)帶即為脂質(zhì)筏。AD組血小板脂質(zhì)筏內(nèi)蛋白含量略高于對(duì)照組,但差異無(wú)統(tǒng)計(jì)學(xué)意義(P=0.1494)。AD組血小板脂質(zhì)筏內(nèi)神經(jīng)節(jié)苷脂GM1含量明顯高于對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P0.01),但其與年齡、MMSE評(píng)分無(wú)相關(guān)性。結(jié)論 AD組患者血小板脂質(zhì)筏內(nèi)神經(jīng)節(jié)苷脂GM1含量與對(duì)照組相比明顯升高,故血小板脂質(zhì)筏內(nèi)神經(jīng)節(jié)苷脂GM1含量的改變?cè)贏D的診斷中有一定的潛在價(jià)值,有望成為一種輔助診斷AD的早期生物學(xué)指標(biāo)。
[Abstract]:Objective to study the changes of ganglioside GM1 in platelet lipid rafts in patients with Alzheimer's disease (AD) and to explore a possible early biological marker for AD diagnosis. Methods 42 patients with AD and 45 healthy controls with normal cognitive function were selected to evaluate their neurological function. Platelet lipid rafts were separated by density gradient centrifugation using Optiprep as the medium. The content of platelet lipid raft protein was determined by BCA method, and the content of ganglioside GM1 was measured by colorimetry and dot blot quantitative analysis. Results A white shiny band was observed at the interface of 25%~30%Optiprep solution in centrifuge tube. The white shiny band located in the sixth layer was identified as lipid raft by dot blotting. The protein content of platelet lipid raft in AD group was slightly higher than that in control group. But there was no significant difference (P < 0. 1494). AD). The content of ganglioside GM1 in platelet lipid raft was significantly higher than that in control group (P0. 01), but it had no correlation with age and MMSE score. Conclusion the content of ganglioside GM1 in platelet lipid raft in AD group is significantly higher than that in control group, so the change of GM1 content in platelet lipid raft may have some potential value in the diagnosis of AD. It is expected to be an early biological marker for auxiliary diagnosis of AD.
