【摘要】：目的研究阿爾茨海默病(AD)患者血小板脂質筏內神經(jīng)節(jié)苷脂GM1含量的變化,尋求一種可能作為輔助診斷AD的早期生物學指標。方法選擇42例AD患者和45例認知功能正常的健康對照者分別進行神經(jīng)功能評定,采用以Optiprep為介質的密度梯度離心方法分離血小板脂質筏,應用點印跡的方法進行鑒定后,采用BCA法測定血小板脂質筏內蛋白含量,比色法及點印跡定量分析法測定其內神經(jīng)節(jié)苷脂GM1含量。結果在離心管25%~30%Optiprep溶液界面處可見一條白色的閃亮環(huán)帶,經(jīng)點印跡法證實,位于第6層的此白色閃亮環(huán)帶即為脂質筏。AD組血小板脂質筏內蛋白含量略高于對照組,但差異無統(tǒng)計學意義(P=0.1494)。AD組血小板脂質筏內神經(jīng)節(jié)苷脂GM1含量明顯高于對照組,差異有統(tǒng)計學意義(P0.01),但其與年齡、MMSE評分無相關性。結論 AD組患者血小板脂質筏內神經(jīng)節(jié)苷脂GM1含量與對照組相比明顯升高,故血小板脂質筏內神經(jīng)節(jié)苷脂GM1含量的改變在AD的診斷中有一定的潛在價值,有望成為一種輔助診斷AD的早期生物學指標。
[Abstract]:Objective to study the changes of ganglioside GM1 in platelet lipid rafts in patients with Alzheimer's disease (AD) and to explore a possible early biological marker for AD diagnosis. Methods 42 patients with AD and 45 healthy controls with normal cognitive function were selected to evaluate their neurological function. Platelet lipid rafts were separated by density gradient centrifugation using Optiprep as the medium. The content of platelet lipid raft protein was determined by BCA method, and the content of ganglioside GM1 was measured by colorimetry and dot blot quantitative analysis. Results A white shiny band was observed at the interface of 25%~30%Optiprep solution in centrifuge tube. The white shiny band located in the sixth layer was identified as lipid raft by dot blotting. The protein content of platelet lipid raft in AD group was slightly higher than that in control group. But there was no significant difference (P < 0. 1494). AD). The content of ganglioside GM1 in platelet lipid raft was significantly higher than that in control group (P0. 01), but it had no correlation with age and MMSE score. Conclusion the content of ganglioside GM1 in platelet lipid raft in AD group is significantly higher than that in control group, so the change of GM1 content in platelet lipid raft may have some potential value in the diagnosis of AD. It is expected to be an early biological marker for auxiliary diagnosis of AD.
