產(chǎn)前應(yīng)激孕鼠子代抑郁樣行為觀察及機(jī)制探討

【摘要】:目的觀察產(chǎn)前應(yīng)激孕鼠子代鼠的抑郁樣行為,并探討產(chǎn)前應(yīng)激導(dǎo)致子代鼠出現(xiàn)抑郁樣行為的作用機(jī)制。方法將孕鼠20只隨機(jī)分觀察組和對(duì)照組各10只。妊娠第14天(孕鼠進(jìn)入孕晚期)起對(duì)觀察組給予束縛應(yīng)激,45 min/次,3次/d,共束縛7 d。對(duì)照組孕鼠孕期不做任何處理,常規(guī)飼養(yǎng)。兩組子代鼠出生30 d時(shí)各取20只(雌雄對(duì)半),進(jìn)行糖水偏愛試驗(yàn),計(jì)算兩組雌、雄性子代鼠1%糖水消耗比(1%糖水消耗比表示抑郁性行為嚴(yán)重程度);隨之將試驗(yàn)后子代鼠斷頭處死,采用液相色譜串聯(lián)質(zhì)譜分析法檢測(cè)兩組子代鼠海馬組織谷氨酸(Glu)、γ-氨基丁酸(GABA)和5-羥色胺(5-HT)。結(jié)果觀察組雌、雄性子代鼠1%糖水消耗比分別為63.13±3.47和59.67±3.33,對(duì)照組分別為79.22±7.07和78.67±4.72;觀察組雌、雄性子代鼠1%糖水消耗比與對(duì)照組相比,P均0.05。觀察組雌、雄子代鼠海馬組織Glu的相對(duì)含量分別為(24.30±1.00)、(31.22±1.57)ng/g,對(duì)照組分別為(21.20±0.90)、(24.01±0.28)ng/g,觀察組雌、雄性子代鼠海馬組織Glu的相對(duì)含量與對(duì)照組相比,P均0.05;觀察組雌、雄子代鼠海馬組織GABA的相對(duì)含量分別為(7.42±0.36)、(8.35±0.39)ng/g,對(duì)照組分別為(13.28±0.38)、(9.94±0.30)ng/g,觀察組雌、雄性子代鼠海馬組織GABA的相對(duì)含量與對(duì)照組相比,P均0.05;觀察組雌、雄子代鼠海馬組織5-HT的相對(duì)含量分別為(0.27±0.02)、(0.26±0.01)ng/g,對(duì)照組分別為(0.35±0.03)、(0.29±0.02)ng/g,觀察組雌性子代大鼠海馬組織5-HT的相對(duì)含量與對(duì)照組相比,P0.05。結(jié)論產(chǎn)前應(yīng)激可導(dǎo)致子代鼠出現(xiàn)抑郁樣行為,其機(jī)制可能與產(chǎn)前應(yīng)激升高子代鼠海馬組織Glu含量及降低GABA、5-HT的水平有關(guān)。
[Abstract]:Objective to observe the depression-like behavior of offspring of pregnant rats under prenatal stress and to explore the mechanism of depressive behavior induced by prenatal stress. Methods Twenty pregnant rats were randomly divided into observation group (n = 10) and control group (n = 10). From the 14th day of pregnancy (the pregnant rats entered the third trimester of pregnancy), the observation group was given restraint stress for 45 min/ 3 / d for a total of 7 days. The pregnant rats in the control group did not do any treatment during pregnancy. At 30 days after birth, 20 offspring of each group (female and male half) were selected for sugar water preference test, and the two groups of females were calculated. The male offspring had a ratio of 1% sugar to water consumption (1% sugar water consumption ratio indicated the severity of depressive behavior), and then the offspring were killed after the experiment. (Glu), 緯 -aminobutyric acid (GABA) and serotonin (5-HT) in hippocampus of two offspring rats were determined by liquid chromatography tandem mass spectrometry (LC-MS). Results the ratio of 1% sugar to water consumption of female and male offspring was 63.13 鹵3.47 and 59.67 鹵3.33 in the observation group and 79.22 鹵7.07 and 78.67 鹵4.72 in the control group, respectively. The relative content of Glu in hippocampal tissue of female and male offspring of observation group was (24.30 鹵1.00), (31.22 鹵1.57) ng/g, control group was (21.20 鹵0.90), () 24.01 鹵0.28 ng/g, group, the relative content of Glu in hippocampal tissue of male offspring group was 0.05 compared with that of control group (P < 0.05). The relative content of GABA in hippocampal tissue of male offspring rats was (7.42 鹵0.36), (8.35 鹵0.39) ng/g, control group was (13.28 鹵0.38), (9.94 鹵0.30) ng/g, observation group, and the relative content of GABA in hippocampal tissue of male offspring rats was 0.05 compared with that of control group (P < 0.05). The relative content of 5-HT in hippocampus of male offspring was (0.27 鹵0.02), (0.26 鹵0.01) ng/g, control group was (0.35 鹵0.03), () 0.29 鹵0.02 ng/g, observation group, the relative content of 5-HT in hippocampus of female offspring group was higher than that of control group (P 0.05). Conclusion Prenatal stress may lead to depressive behavior in offspring, and the mechanism may be related to the increase of Glu content in hippocampus and the decrease of GABA,5-HT in hippocampus.
