【摘要】：目的研究氯胺酮長期濫用模型中海馬區(qū)半胱氨酸蛋白酶(caspase)3和突觸后密度蛋白(PSD)95的表達變化。方法每日小鼠腹腔注射氯胺酮(30 mg/kg)、持續(xù)3個月建立小鼠氯胺酮長期濫用模型,采用免疫組化及Western blotting檢測小鼠氯胺酮長期濫用后caspase-3和PSD-95的表達情況。結果免疫組化結果顯示caspase-3表達增強,PSD-95表達減弱。Western blotting結果顯示caspase-3活化片段蛋白表達升高,PSD-95蛋白表達下降。結論氯胺酮長期濫用致小鼠腦海馬區(qū)caspase-3表達增加、PSD-95表達減弱,這一結果揭示了長期濫用氯胺酮所致神經毒性作用的可能機制。
[Abstract]:Objective to study the expression of cysteine protease (caspase) 3 and postsynaptic density protein (PSD) 95 in hippocampus of ketamine model. Methods Ketamine (30 mg/kg) was injected intraperitoneally daily to establish the model of long-term abuse of ketamine in mice for 3 months. The expression of caspase-3 and PSD-95 were detected by immunohistochemistry and Western blotting. Results Immunohistochemical results showed that the expression of caspase-3 increased and the expression of PSD-95 decreased. Western blotting showed that the expression of caspase-3 activated fragment protein increased and the expression of PSD-95 decreased. Conclusion the long-term abuse of ketamine can increase the expression of caspase-3 and decrease the expression of PSD-95 in the hippocampus of mice, which reveals the possible mechanism of neurotoxicity induced by long-term abuse of ketamine.
