【摘要】：目的:探討組織轉(zhuǎn)谷氨酰胺酶2(TGM2)基因多態(tài)性與中國北方漢族人群精神分裂癥發(fā)病的關(guān)系,闡明精神分裂癥發(fā)病的遺傳因素。方法:收集428例中國北方漢族人群精神分裂癥患者和555名健康對照者全血樣品,提取基因組DNA,采用聚合酶鏈式反應(yīng)-限制性片段長度多態(tài)性(PCR-RFLP)的方法檢測TGM2基因rs2076380、rs7270785、rs4811528和rs6023526位點基因型。應(yīng)用擬合優(yōu)度χ2檢驗分析4個單核苷酸多態(tài)性(SNPs)位點基因型頻數(shù)分布是否符合Hardy-Weinberg平衡定律,應(yīng)用χ2檢驗和數(shù)量性狀分析分別進行等位基因、基因型的關(guān)聯(lián)性分析。結(jié)果:TGM2基因rs2076380、rs7270785、rs4811528和rs6023526位點的基因型頻數(shù)分布在精神分裂癥病例組和健康對照組中均符合Hardy-Weinberg平衡(P0.05)。等位基因關(guān)聯(lián)分析,病例組和對照組中各SNPs位點等位基因的頻數(shù)分布比較差異無統(tǒng)計學(xué)意義(均P0.05)。基因型關(guān)聯(lián)分析,病例組和對照組中各SNPs位點基因型的頻數(shù)分布比較差異無統(tǒng)計學(xué)意義(均P0.05)。臨床表型分析,各SNPs位點與精神分裂癥陽性、陰性臨床表型無關(guān)聯(lián)(P0.05)。結(jié)論:TMG2基因rs2076380、rs7270785、rs4811528和rs6023526位點多態(tài)性與中國北方漢族人群精神分裂癥發(fā)病無關(guān)聯(lián)。
[Abstract]:Objective: to investigate the relationship between the polymorphism of tissue transglutaminase 2 (TGM2) gene and the onset of schizophrenia in Han population of northern China, and to elucidate the genetic factors of schizophrenia. Methods: the blood samples of 428 schizophrenic patients and 555 healthy controls were collected. Genomic DNA was extracted and the genotypes of TGM2 gene rs2076380, rs7270785, rs4811528 and rs6023526 were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The genotype frequency distribution of four single nucleotide polymorphisms (SNPs) loci was analyzed by 蠂 ~ 2 test of fit, and the correlation of alleles and genotypes was analyzed by 蠂 ~ 2 test and quantitative trait analysis, respectively. Results the genotypic frequencies of rs2076380 rs7270785rs4811528 and rs6023526 locus of the 1: TGM2 gene were in accordance with the Hardy-Weinberg balance in schizophrenia and healthy controls (P0.05). Allelic association analysis showed that there was no significant difference in the frequency distribution of alleles at each SNPs locus between case group and control group (P0.05). Genotypic association analysis showed that there was no significant difference in the frequency distribution of SNPs loci between the case group and the control group (P0.05). Clinical phenotypic analysis, each SNPs locus and schizophrenia positive, negative clinical phenotypes were not correlated (P0.05). Conclusion there is no association between the polymorphism of rs7270785rs4811528 and rs6023526 and the prevalence of schizophrenia in Han population of northern China.
