【摘要】：目的雷帕霉素可通過(guò)提高自噬水平,改善阿爾茨海默病特征性病理,但其內(nèi)在機(jī)制尚需進(jìn)一步研究。文中旨在探討雷帕霉素對(duì)β淀粉樣蛋白25-35(Aβ25-35)誘導(dǎo)PC12細(xì)胞損傷的保護(hù)作用機(jī)制。方法取對(duì)數(shù)生長(zhǎng)期細(xì)胞PC12分為5組:對(duì)照組(等量無(wú)血清DMEM)、模型組及10μmol/L雷帕霉素組、40μmol/L雷帕霉素組、160μmol/L雷帕霉素組(分別用加入10、40、160μmol/L的雷帕霉素),除對(duì)照組外,其余各組均用20μmol/L的Aβ25-35作用于PC12細(xì)胞構(gòu)建細(xì)胞損傷模型。結(jié)果 (1)與模型組細(xì)胞存活率[(51.47±2.59)%]、凋亡率[(52.22±2.33)%]比較,10、40、160μmol/L雷帕霉素組、對(duì)照組細(xì)胞存活率[(54.64±2.42)、(64.79±2.91)、(56.50±2.55)、(99.98±0.73)%]明顯升高,凋亡率[(45.33±2.83)、(36.89±2.85)、(48.00±2.83)、(3.33±2.45)%]明顯降低(P0.05)。(2)模型組p-PKB表達(dá)量為0.33±0.01,p-m TOR表達(dá)量為1.97±0.05,p-tau表達(dá)量為2.09±0.19;與模型組上述指標(biāo)比較,10、40、160μmol/L雷帕霉素組、對(duì)照組p-PKB表達(dá)量(0.37±0.01、0.42±0.01、0.40±0.01、0.44±0.02)明顯升高(P0.05),而p-m TOR表達(dá)量(1.64±0.05、0.66±0.04、0.35±0.01、0.62±0.01)和p-tau表達(dá)量(2.02±0.15、1.79±0.05、1.86±0.06、1.53±0.04)明顯降低(P0.05)。結(jié)論雷帕霉素能提高Aβ25-35損傷的PC12細(xì)胞的存活率,降低細(xì)胞凋亡率,對(duì)Aβ25-35致PC12細(xì)胞損傷有保護(hù)作用。其機(jī)制可能與雷帕霉素抑制p-m TOR,負(fù)反饋調(diào)節(jié)PI3K/PKB/m TOR信號(hào)轉(zhuǎn)導(dǎo)通路,減少tau蛋白過(guò)度磷酸化有關(guān)。
[Abstract]:Objective rapamycin can improve the characteristic pathology of Alzheimer's disease by increasing autophagy, but its intrinsic mechanism needs further study. The aim of this study was to investigate the protective mechanism of rapamycin on 尾 -amyloid 25-35 (A 尾 25-35) -induced injury in PC12 cells. Methods PC12 cells in logarithmic growth phase were divided into 5 groups: control group (equal dose without serum DMEM), model group and 10 渭 mol / L rapamycin group (40 渭 mol / L rapamycin group, 160 渭 mol / L rapamycin group). The other groups were treated with A 尾 25-35 at 20 渭 mol / L for PC12 cells.
