【摘要】:目的:探討中國(guó)漢族群體5-羥色胺2C受體基因(5-HTR2C)啟動(dòng)區(qū)相關(guān)單核苷酸多態(tài)(SNP)位點(diǎn)rs518147多態(tài)性的分布狀況及其與強(qiáng)迫癥易感性之間的關(guān)系。 方法:采用直接測(cè)序法分析中國(guó)漢族群體符合中國(guó)精神障礙分類(lèi)與診斷標(biāo)準(zhǔn)第3版或國(guó)際疾病分類(lèi)第10版強(qiáng)迫癥診斷標(biāo)準(zhǔn)的患者308例(強(qiáng)迫癥組:男123例,女185例)和410名健康對(duì)照(對(duì)照組:男175例,女235例)5-HTR2Crs518147位點(diǎn)半合子(男性)或基因型(女性)分布頻率,并分析半合子(男性)或基因型(女性)分布頻率與強(qiáng)迫癥發(fā)病的遺傳易感性的關(guān)系。 結(jié)果:強(qiáng)迫癥組中5-HTR2C啟動(dòng)區(qū)rs518147位點(diǎn)男性患者攜帶G半合子與女性患者攜帶G+(GG與GC)基因型的比率均顯著高于對(duì)照組(男性:x2=4.973,P=0.026;女性:x2=5.243,P=0.022),G等位基因的頻率明顯高于對(duì)照組(x2=4.611,P=0.032)。 結(jié)論:中國(guó)漢族群體中5-HTR2C啟動(dòng)區(qū)rs518147位點(diǎn)多態(tài)性可能與強(qiáng)迫癥遺傳易感性相關(guān)。
[Abstract]:Objective: to investigate the distribution of rs518147 polymorphism in the promoter region of serotonin 2C receptor gene (5-HTR2C) and its relationship with susceptibility to obsessive-compulsive disorder (OCD) in Chinese Han population. Methods: a total of 308 patients with obsessive-compulsive disorder (OCD group: 123 males) who met the criteria for the classification and diagnosis of mental disorders in China or the 10th edition of the International Classification of Diseases were analyzed by direct sequencing. The frequencies of heminzygote (male) or genotype (female) of 5-HTR2Crs518147 locus were found in 185 females and 410 healthy controls (control group: 175 males and 235 females). The relationship between the frequency of heminzygote (male) or genotype (female) and the genetic susceptibility of obsessive-compulsive disorder (OCD) was analyzed. Results: in the OCD group, the ratio of G heminzygote and G GG / GC genotype in male patients with 5-HTR2C promoter region was significantly higher than that in the control group (male: x2n 4.973), and in female, the frequency of G allele was significantly higher than that in the control group (male: x25.243), and the frequency of G allele in female was significantly higher than that in the control group (x24.611PU 0.032). Conclusion: the polymorphism of 5-HTR2C rs518147 locus in Chinese Han population may be related to the genetic susceptibility of OCD.
