【摘要】：目的探討Aβ3-10重復(fù)片段質(zhì)粒免疫接種3月齡APPswe/PSEN1雙轉(zhuǎn)基因(AD)鼠腦內(nèi)BACE1的影響。方法應(yīng)用Aβ3-10重復(fù)片段質(zhì)粒免疫3月齡APPswe/PSEN1雙轉(zhuǎn)基因鼠,同等劑量的PBS免疫對(duì)照組及C57BL/6J組小鼠,各組小鼠均在免疫后2、4、6次后通過RT-PCR法檢測(cè)BACE1 m RNA水平,Western blotting法檢測(cè)BACE1/GFAP/NF-κB蛋白水平,免疫組化法觀察Aβ1-42腦內(nèi)分布情況。水迷宮檢測(cè)其行為學(xué)改變。結(jié)果在免疫2次后BACE1 m RNA表達(dá)水平C57BL/6J組Aβ3-10組對(duì)照組(F=4.649,P=0.021);免疫4次Aβ3-10組C57BL/6J組對(duì)照組(F=115.683,P=0.001);免疫6次C57BL/6J組Aβ3-10組對(duì)照組(F=86.600,P0.001)。BACE1的蛋白表達(dá)水平在免疫2、4、6次后C57BL/6J組Aβ3-10組對(duì)照組(F=432.843,P0.001;F=57.673,P0.001;F=26.550,P=0.001),NF-κB的蛋白表達(dá)水平在免疫2次后C57BL/6J組Aβ3-10組對(duì)照組(F=109.127,P0.001);免疫4,6次后Aβ3-10組C57BL/6J組對(duì)照組(F=30.301,P0.001;F=129.967,P0.001)。GFAP的蛋白表達(dá)水平在免疫2、4、6次后C57BL/6J組Aβ3-10組對(duì)照組(F=27.782,P=0.001;F=26.132,P=0.001;F=26.450,P=0.001);Aβ1-42在腦內(nèi)的分布C57BL/6J組Aβ3-10組對(duì)照組(皮質(zhì):F=5.395,P=0.021;F=47.135,P=0.000;F=25.306,P=0.000,海馬:F=11.023,P=0.002;F=14.936,P=0.001;F=50.132,P=0.000)。總潛伏期C57BL/6J組Aβ3-10組對(duì)照組(F=8.938,P=0.016;F=5.745,P=0.04;F=7.073,P=0.017)。結(jié)論 Aβ3-10重復(fù)片段質(zhì)粒可以影響腦內(nèi)BACE1的表達(dá),影響Aβ產(chǎn)生,改善空間記憶能力。
[Abstract]:Objective to investigate the effect of A 尾 3-10 repeat plasmid immunization on BACE1 in the brain of 3 month old APPswep / PSEN1 transgenic mice. Methods A 尾 3-10 repeat fragment plasmid was used to immunize 3-month-old APPswe-PSEN1 double transgenic mice. The control group and C57BL / 6J group were immunized with the same dose of PBS. The BACE1 mRNA level was detected by RT-PCR and the protein level of BACE1 / GFAPNF- 魏 B was detected by RT-PCR. The distribution of A 尾 1-42 in brain was observed by immunohistochemical method.
