氟西汀對人嗅黏膜神經(jīng)干細(xì)胞增殖與神經(jīng)發(fā)生的影響

【摘要】:目的探索以氟西汀為代表的抗抑郁藥對人嗅黏膜神經(jīng)干細(xì)胞增殖及神經(jīng)發(fā)生的影響。為抗抑郁治療能夠促進(jìn)神經(jīng)發(fā)生這一理論增添新的證據(jù)。方法采用組織塊培養(yǎng)法獲取人嗅黏膜細(xì)胞后,用神經(jīng)球培養(yǎng)法純化擴(kuò)增神經(jīng)干細(xì)胞,采用細(xì)胞免疫熒光染色驗(yàn)證神經(jīng)干細(xì)胞及由其分化而來的神經(jīng)細(xì)胞的特異性。分別采用CCK-8、Edu技術(shù)檢測抗抑郁藥對嗅黏膜神經(jīng)干細(xì)胞活性及增殖的影響;采用蛋白質(zhì)分子印記技術(shù)檢測抗抑郁藥對其神經(jīng)發(fā)生的影響。結(jié)果1.Nestin(神經(jīng)干細(xì)胞標(biāo)志蛋白)細(xì)胞熒光染色陽性;由其分化而來的細(xì)胞TUJ1(神經(jīng)細(xì)胞特異標(biāo)志物)細(xì)胞熒光染色陽性。神經(jīng)干細(xì)胞培養(yǎng)成功。2.CCK-8實(shí)驗(yàn)顯示一定濃度的氟西汀可加強(qiáng)嗅黏膜神經(jīng)干細(xì)胞的活力;Edu實(shí)驗(yàn)顯示一定濃度的氟西汀可促進(jìn)神經(jīng)干細(xì)胞增殖。3.蛋白質(zhì)分子印跡實(shí)驗(yàn)顯示一定濃度氟西汀處理的嗅黏膜神經(jīng)干細(xì)胞,其TUJ1的表達(dá)量較對照組顯著上升,即氟西汀可以促進(jìn)干細(xì)胞分化成神經(jīng)細(xì)胞。結(jié)論1.人嗅黏膜神經(jīng)干細(xì)胞原代培養(yǎng)成功且其可分化為神經(jīng)細(xì)胞。2.抗抑郁治療可促進(jìn)神經(jīng)干細(xì)胞的增殖及向神經(jīng)細(xì)胞分化,有可能為抗抑郁藥物治療的機(jī)制之一。
[Abstract]:Objective to investigate the effects of fluoxetine on the proliferation and neurogenesis of neural stem cells in human olfactory mucosa. Add new evidence to the theory that antidepressant therapy promotes neurogenesis. Methods Human olfactory mucosal cells were obtained by tissue mass culture. The neural stem cells were purified and amplified by the method of neurosphere culture. The specificity of neural stem cells and the nerve cells derived from them were tested by immunofluorescence staining. CCK-8 Edu technique was used to detect the effects of antidepressants on the activity and proliferation of neural stem cells in olfactory mucosa, and protein molecular imprinting technique was used to detect the effects of antidepressants on neurogenesis. Results 1. Nestin (neural stem cell marker protein) cells were positive for fluorescence staining, and TUJ1 (neuronal specific marker) cells from Nestin (neural stem cell marker protein) cells were positive for fluorescence staining. CCK-8 experiment showed that a certain concentration of fluoxetine could enhance the activity of neural stem cells in olfactory mucosa. Edu experiment showed that a certain concentration of fluoxetine could promote the proliferation of neural stem cells. Western blot analysis showed that the expression of TUJ1 in olfactory mucosal neural stem cells treated with fluoxetine at a certain concentration was significantly higher than that in control group, that is, fluoxetine could promote the differentiation of stem cells into neural cells. Conclusion 1. Human olfactory mucosal neural stem cells were successfully cultured and differentiated into neural cells. 2. Antidepressant therapy can promote the proliferation and differentiation of neural stem cells, which may be one of the mechanisms of antidepressant therapy.
