雌激素對(duì)去卵巢大鼠抑郁樣行為及杏仁核NGF表達(dá)的影響

【摘要】:目的通過觀察雌二醇治療對(duì)雌激素剝奪大鼠的抑郁樣行為及杏仁核神經(jīng)生長因子(NGF)含量的變化,探討雌二醇治療抑郁癥的作用靶點(diǎn)及作用機(jī)制。方法健康成年雌性SD大鼠隨機(jī)分為假手術(shù)組(SHAM組,n=10)、卵巢切除組(OVX組,n=10)及去卵巢雌二醇治療組(OVX+E組,n=10)3組。雌二醇治療8周后通過懸尾及糖水偏愛實(shí)驗(yàn)觀察雌二醇對(duì)雌激素剝奪大鼠行為學(xué)的影響;隨后用免疫組織化學(xué)方法測(cè)量和分析杏仁核NGF神經(jīng)元的數(shù)量及面積。結(jié)果與SHAM組大鼠比較,OVX組大鼠糖水偏愛實(shí)驗(yàn)中糖水偏愛率顯著下降(P0.01)、懸尾實(shí)驗(yàn)中不動(dòng)時(shí)間延長(P0.01),血清雌二醇水平及杏仁核NGF表達(dá)均顯著下降(P均0.01);8周雌二醇治療改善了去卵巢大鼠的抑郁樣行為,與OVX組大鼠比較,血清雌激素水平及杏仁核NGF表達(dá)水平均顯著增加(P均0.01)。結(jié)論雌二醇治療改善了去卵巢大鼠的抑郁樣行為,可能與雌二醇增加去卵巢大鼠的血清雌激素水平及增強(qiáng)杏仁核NGF的表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effects of estradiol on depression and the changes of nerve growth factor (NGF) content in amygdaloid nucleus of estrogen deprived rats. Methods healthy adult female Sprague-Dawley rats were randomly divided into three groups: sham-operated group (sham group), ovariectomized group (OVX group) and ovariectomized estradiol group (OVX E group). After 8 weeks of estradiol treatment, the effects of estradiol on the behavior of estrogen deprived rats were observed by tail suspension and sugar water preference experiments, and the number and area of NGF neurons in amygdala were measured and analyzed by immunohistochemical method. Results compared with SHAM group, the rate of glucose water preference was significantly decreased in OVX group, and the immobility time was prolonged in tail suspension test. The serum estradiol level and NGF expression in amygdala were significantly decreased in OVX group after 8 weeks of estradiol treatment. Improved depressive behavior in ovariectomized rats, Compared with OVX group, the level of serum estrogen and the expression of NGF in amygdala increased significantly (P < 0.01). Conclusion Estradiol therapy can improve the depressive behavior of ovariectomized rats, which may be related to the increase of serum estrogen level and the increase of amygdaloid NGF expression in ovariectomized rats.
