大鼠創(chuàng)傷性腦損傷后精神障礙的精神行為學(xué)改變

【摘要】:目的利用改良FEENEY’S法建立大鼠創(chuàng)傷性腦損傷(TBI)模型,探討TBI后精神障礙的精神行為學(xué)改變。方法將成年雄性SD大鼠隨機(jī)分為正常對(duì)照組、TBI組和假手術(shù)組,每組各10只。采用改良FEENEY’S法建立大鼠TBI模型。TBI后15 d開始,分別依次進(jìn)行曠場(chǎng)實(shí)驗(yàn)(OFT)和強(qiáng)迫游泳實(shí)驗(yàn)(FST)。記錄OFT中大鼠5 min內(nèi)的移動(dòng)總距離(DMT)、平均速度(VM),記錄FST中大鼠5 min內(nèi)不動(dòng)時(shí)間總和(ITT),觀察大鼠的高警覺性、焦慮/抑郁和恐懼行為的變化。結(jié)果 TBI組大鼠的DMT[(1 228.10±204.97)cm]、運(yùn)動(dòng)VM[(4.09±0.68)cm/s]較假手術(shù)組[(1 660.01±332.09)cm、(5.53±1.10)cm/s]和正常對(duì)照組[(2 022.32±280.92)cm、(7.28±1.80)cm/s]均明顯減少,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。TBI組大鼠的ITT[(149.88±27.36)s]較假手術(shù)組[(120.55±22.31)s]、正常對(duì)照組[(106.10±29.03)s]明顯增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 TBI后,大鼠出現(xiàn)了焦慮/抑郁樣精神行為異常。
[Abstract]:Objective to establish a model of traumatic brain injury (TBI) in rats with modified FEENEY'S method and to investigate the changes of mental disorders after TBI. Methods Adult male Sprague-Dawley rats were randomly divided into normal control group (TBI) and sham operation group (10 rats in each group). The rat model of TBI was established by modified FEENEY'S method. The open field experiment and forced swimming test were carried out 15 days later. The total distance and average velocity of OFT rats within 5 min were recorded, and the total immobility time of 5 min in FST rats were recorded.
