Znf804a基因在發(fā)育期鼠腦神經(jīng)元的遷移、增殖及分化中的功能

【摘要】:目的:以小鼠為模型研究精神分裂癥易感基因Znf804a在神經(jīng)發(fā)育中的功能。方法:利用胚胎電轉(zhuǎn)技術(shù),將Znf804a的shRNA(shZnf804a)質(zhì)粒及其對照質(zhì)粒(pSUPER)電轉(zhuǎn)進(jìn)入E14.5的癌癥研究所(ICR)小鼠腦室區(qū),每種質(zhì)粒電轉(zhuǎn)3只,分別為敲低(knock down)Znf804a組和對照組,比較兩組神經(jīng)元在皮層板(CP)區(qū)的比例來評價(jià)神經(jīng)元的遷移速度;比較兩組神經(jīng)元前體細(xì)胞形成神經(jīng)球的直徑來評估神經(jīng)元增殖速度;比較體外培養(yǎng)神經(jīng)元前體細(xì)胞中Nestin染色陽性神經(jīng)元的比例檢測神經(jīng)元分化速度。結(jié)果:敲低Znf804a小鼠的神經(jīng)元遷移到CP區(qū)的比例比對照組低(11.8%vs.75.4%,P0.001);神經(jīng)元前體細(xì)胞形成神經(jīng)球的直徑比對照組大(295μm vs.172μm,P0.01);神經(jīng)元前體細(xì)胞中,Nestin染色陽性的神經(jīng)元比例比對照組高(31.5%vs.9.6%,P0.01)。結(jié)論:敲低Znf804a小鼠神經(jīng)元遷移速度和分化速度比對照組神經(jīng)元慢,增殖速度比對照組神經(jīng)元快,提示Znf804a在小鼠腦發(fā)育過程中發(fā)揮重要功能。
[Abstract]:Objective: to study the function of susceptibility gene Znf804a in nervous development in mice. Methods: the shRNA-shZnf804a) plasmid of Znf804a and its control plasmid pSUPERA were electrotransferred into the ventricular region of the mice of cancer research institute of E14.5 by electroporation technique. Each plasmid was electrotransferred into 3 mice, which were knock down knock down)Znf804a group and control group, respectively. The migration rate of neurons was evaluated by comparing the proportion of neurons in the cortex (CPV) region, the diameter of neuronal precursor cells forming neurospheres to evaluate the proliferation rate of neurons in the two groups. The rate of Nestin staining positive neurons in cultured neurons in vitro was compared. Results: the percentage of neurons migrating to CP area in Znf804a knockout mice was 11.8vs.75.4i P0.001, the diameter of neuronal precursor cells was larger than that of control group (295 渭 m vs.172 渭 m P0.01), and the percentage of nestin positive neurons in neurons precursor cells was higher than that in control group (31.5 vs.9.6m P0.01). Conclusion: the migration and differentiation of neurons in Znf804a knockout mice are slower than those in control group, and the proliferation rate is faster than that in control group, suggesting that Znf804a plays an important role in the development of mouse brain.
