海馬髓鞘堿性蛋白表達的改變對大鼠抑郁樣行為的影響

【摘要】:抑郁癥是危害人類身心健康的一種常見的精神類疾病。近年來,隨著社會的進步,人們的生活節(jié)奏也越來越快,抑郁癥的發(fā)病率也在不斷呈現(xiàn)上升的趨勢。 髓鞘堿性蛋白(myelin basic protein,MBP)是脊椎動物中樞神經(jīng)系統(tǒng)(central nerve system, CNS)少突膠質(zhì)細胞和周圍神經(jīng)系統(tǒng)雪旺細胞合成的一種弱堿性膜蛋白,含有多種堿性氨基酸。MBP是中樞神經(jīng)系統(tǒng)(CNS)髓鞘的主要蛋白質(zhì),位于髓鞘漿膜面,維持CNS髓鞘結(jié)構(gòu)和功能的穩(wěn)定,具有神經(jīng)組織特異性。 目的:探討MBP蛋白表達的改變對抑郁大鼠的行為活動的影響 方法:大鼠隨機分為正常組、模型組、MBP高表達組、RNA干擾組和空載體組。應用慢性不可預知溫和應激刺激制備抑郁大鼠模型。應用強迫游泳實驗,高架十字迷宮實驗,曠場實驗等行為學測試方法觀察MBP對抑郁大鼠行為活動的影響。用免疫熒光觀察MBP的表達,用免疫組化的方法觀察MBP的分布,用Western Blot法檢測MBP的含量。 結(jié)果: 1.MBP對CUS大鼠抑郁樣行為的影響 從強迫游泳實驗中可以看出,造模8周模型組大鼠的不動時間與正常組相比有顯著性差異(P0.05)。誘導MBP表達后,MBP高表達組大鼠的不動時間比模型組大鼠的不動時間縮短,差異有統(tǒng)計學意義(P0.05)?蛰d體組和RNA干擾組相對于模型組來說無顯著性差異。 從高架十字迷宮的實驗中可以看出,造模8周模型組大鼠的開臂滯留時間百分比比正常組下降,差異有統(tǒng)計學意義(P0.05)。誘導MBP表達后,MBP高表達組大鼠的開臂滯留時間百分比比模型組延長,差異有統(tǒng)計學意義(P0.05)?蛰d體組和RNA干擾組相對于模型組來說無顯著性差異。 在曠場實驗中,從水平方向運動情況看,造模8周模型組大鼠的水平總位移比正常組大鼠下降,差異有統(tǒng)計學意義(P0.05)。誘導MBP表達后,MBP高表達組大鼠的水平總位移相對于模型組來說升高,差異有統(tǒng)計學意義(P0.05),但仍小于正常組?蛰d體組和RNA干擾組相對于模型組來說無顯著性差異。從垂直方向的直立次數(shù)看,造模8周模型組大鼠的直立次數(shù)減少,與正常組比有顯著性差異(P0.05)。誘導MBP表達后,MBP高表達組大鼠的直立次數(shù)相對于模型組來說增加,差異有統(tǒng)計學意義(P0.05),空載體組和RNA干擾組相對于模型組來說無顯著性差異。 2.大鼠海馬MBP的變化 免疫熒光中看出,熒光強度由高到低依次為MBP高表達組,空載體組,RNA干擾組。正常組和模型組中無熒光。 免疫組化實驗看出MBP高表達組比模型組MBP蛋白表達要多,差異有統(tǒng)計學意義(P0.05),RNA干擾組比模型組MBP蛋白表達要少,差異有統(tǒng)計學意義(P0.05)。 Western Blot檢測得出,MBP高表達組大鼠海馬MBP蛋白含量顯著高于模型組,差異有統(tǒng)計學意義(P0.05),RNA干擾組大鼠海馬MBP蛋白含量顯著低于高表達組,差異有統(tǒng)計學意義(P0.05)。 結(jié)論: 1.通過誘導MBP基因的改變可以引起其蛋白表達的改變 2.MBP蛋白表達水平的改變可以引起抑郁樣行為的變化
[Abstract]:Depression is a common mental disease which endangers the physical and mental health of human beings. In recent years, with the progress of the society, people's life rhythm is getting faster and faster, and the incidence of depression is also increasing.
Myelin basic protein (MBP) is a weak alkaline membrane protein synthesized from the central nervous system of vertebrate (central nerve system, CNS) oligodendrocytes and peripheral nervous system Schwann cells. It contains a variety of basic amino acids.MBP is the main protein of the myelin sheath in the central nervous system (CNS). It is located in the myelin serous surface and is maintained. CNS myelin sheath is stable in structure and function.
Objective: To investigate the effect of MBP protein expression on behavioral behavior in depressed rats.
Methods: rats were randomly divided into normal group, model group, MBP high expression group, RNA interference group and no-load group. Chronic unpredictable mild stress stimulation was used to prepare depressive rats model. Forced swimming test, elevated cross maze test and open field test were used to observe the effect of MBP on behavior of depressive rats. The expression of MBP was observed by fluorescence. The distribution of MBP was observed by immunohistochemistry. The content of MBP was detected by Western Blot.
