探索GalR1在雌性CUMS C57小鼠海馬表達及M617對干細胞增殖的影響

【摘要】:目的:探究GalR1在雌性CUMS C57小鼠海馬表達及GalR1激動劑M617對干細胞增殖的影響。方法:(1)將小鼠分為實驗組及對照組,對照組不給予任何刺激,實驗組給予慢性溫和不可預知應激(CUMS)。通過糖水偏好、懸尾實驗及血清皮質酮比較兩組的抑郁水平;部分小鼠行免疫熒光染色比較兩組海馬DG的增殖情況;另一部分小鼠行qPCR檢測,比較兩組海馬甘丙肽及其受體的表達。(2)行細胞培養(yǎng),傳代細胞給予不同濃度的M617處理3 d,取每組細胞鋪板并加入10μmol/L的Brd U,繼續(xù)孵育4 h后PFA固定,行Brdu免疫熒光染色并比較各組的陽性細胞數。結果:(1)第2、3周的糖水偏好實驗顯示兩組糖水的消耗量未見差異,第4周CUMS組糖水消耗量少于對照組(P0.05);懸尾實驗中,CUMS組靜止不動時間延長(P0.05);CUMS組血清皮質酮水平明顯高于對照組(P0.01)。(2)CUMS組海馬DG區(qū)的Brd U+及Ki67+細胞數明顯少于對照組(P0.05)。(3)q PCR結果顯示CUMS組甘丙肽及GalR1基因表達水平明顯增加(P0.01)。(4)不同濃度M617對干細胞增殖無明顯組間差異。結論:(1)GalR1在雌性CUMS C57小鼠海馬表達增高,可能參與調節(jié)抑郁癥的發(fā)生過程。(2)不同濃度M617對干細胞增殖無明顯影響,提示GalR1可能不參與神經干細胞增殖的調節(jié)。
[Abstract]:Aim: to investigate the expression of GalR1 in the hippocampus of female CUMS C57 mice and the effect of GalR1 agonist M617 on the proliferation of stem cells.Methods mice were divided into experimental group and control group. The control group was given no stimulation and the experimental group was given chronic mild and unpredictable stress.The depression levels of the two groups were compared by sucrose preference, tail suspension test and serum corticosterone. Some mice were stained with immunofluorescence to compare the proliferation of DG in the hippocampus of the two groups. The other part of the mice were tested for qPCR.The expression of glianin and its receptor was compared between the two groups. The cells were treated with M617 at different concentrations for 3 days. The cells in each group were treated with 10 渭 mol/L Brd U for 3 days. The cells were incubated for 4 h and then PFA was fixed.Brdu immunofluorescence staining was performed and the number of positive cells in each group was compared.Results the sugar water preference experiment of the 2nd week showed no difference between the two groups in the consumption of sugar water.At the 4th week, the consumption of sugar water in CUMS group was less than that in control group (P 0.05), and the serum corticosterone level in CUMS group was significantly higher than that in control group (P 0.01). The number of Brd U and Ki67 cells in DG area of hippocampus in CUMS group was significantly lower than that in control group (P0.05.3q PCR).The results showed that there was no significant difference in the proliferation of stem cells induced by M617 at different concentrations of M617 in CUMS group.Conclusion the expression of Gal R1 in the hippocampus of female CUMS C57 mice is increased, and M617 has no obvious effect on stem cell proliferation. It suggests that GalR1 may not be involved in the regulation of neural stem cell proliferation.
