【摘要】：目的探討姜黃素對AD模型小鼠學習記憶能力的影響。方法將80小鼠按隨機原則分四組,對照組、Alcl3組、溶劑組及姜黃素組。Alcl3制作小鼠AD模型,之后給予腹腔注射姜黃素3 d,給藥后14 d水迷宮測試學習記憶能力。免疫染色及western blot方法檢測Aβ42、GFAP和Iba-1β蛋白水平。ELISA方法檢測海馬區(qū)炎癥因子IL-1β,IL-6和TNF-α的水平。結果與對照組比,Alcl3能夠導致逃避潛伏期延長和記憶下降更明顯,說明AD模型成功建立。與Alcl3組和溶劑治療組相比,姜黃素組明顯減少潛伏期及增加穿越次數(shù),而且顯著減少Aβ42表達。相比Alcl3組,姜黃素組的GFAP和Iba-1β以及海馬區(qū)IL-1β、IL-6及TNF-α的指標下降有顯著意義。結論研究表明姜黃素有潛在治療AD的可能,通過下調Alcl致膠質細胞過度活化引起炎癥反應,而提高學習記憶能力。
[Abstract]:Objective to investigate the effect of curcumin on learning and memory ability of AD model mice. Methods 80 mice were randomly divided into four groups: control group, solvent group and curcumin group. After intraperitoneal injection of curcumin for 3 days, the ability of learning and memory was tested by water maze on the 14th day after administration. The levels of A 尾 42G FAP and Iba-1 尾 protein and Iba-1 尾 protein were detected by immunocytochemistry and western blot method. Elisa was used to detect the levels of IL-1 尾 -guan6 and TNF- 偽 in hippocampal area. In the control group, the escape latency was prolonged and the memory decreased more significantly than that in the control group. Compared with Alcl3 group and solvent treatment group, curcumin group significantly reduced latency and increased traversal times, and significantly reduced A 尾 42 expression. Compared with Alcl3 group, curcumin group significantly decreased the expression of A 尾 42, compared with Alcl3 group, curcumin group significantly decreased the expression of A 尾 42. In curcumin group, the indexes of GFAP and Iba-1 尾 and IL-1 尾 IL-6 and TNF- 偽 in hippocampus decreased significantly. Conclusion curcumin has the potential to treat AD by down-regulating the inflammatory response of glial cells induced by Alcl and improving the ability of learning and memory.
