焦慮模型大鼠下丘腦—垂體—腎上腺軸高反應(yīng)性與皮質(zhì)類(lèi)固醇受體關(guān)系的研究

【摘要】:目的:研究在應(yīng)激條件下焦慮模型大鼠下丘腦-垂體-腎上腺(hypothalamus-pituitary-adrenal,HPA)軸呈高反應(yīng)性是與軸本身功能亢進(jìn)還是與皮質(zhì)類(lèi)固醇受體改變相關(guān)。方法:將SD健康雄性大鼠隨機(jī)分為兩組,對(duì)照組(n=20)和模型組(n=26)。采用不可預(yù)知的情緒應(yīng)激方法建立大鼠焦慮模型,對(duì)照組給予正常飼養(yǎng)。每周稱(chēng)量所有大鼠的體重,并在造模結(jié)束后,通過(guò)行為學(xué)實(shí)驗(yàn)檢測(cè)造模是否成功。高架十字迷宮和曠場(chǎng)作為應(yīng)激源刺激大鼠,測(cè)定兩組大鼠血清中大鼠促腎上腺皮質(zhì)激素(ACTH)和皮質(zhì)酮(CORT)水平。用實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)兩組大鼠海馬組織中鹽皮質(zhì)激素受體(mineralocorticoid steroid receptor,MR)和糖皮質(zhì)激素受體(glucocorticoid steroid receptor,GR)m RNA水平及GR在下丘腦、垂體和腎上腺組織中的m RNA表達(dá)水平。蛋白免疫印跡(Western blot)檢測(cè)兩組大鼠海馬組織內(nèi)MR和GR蛋白水平以及GR在下丘腦、垂體和腎上腺組織中的蛋白表達(dá)水平。結(jié)果:(1)與對(duì)照組相比,焦慮模型大鼠體重增長(zhǎng)率顯著降低(p0.01)。(2)高架十字迷宮和曠場(chǎng)實(shí)驗(yàn)結(jié)果表明,與對(duì)照組相比,模型組大鼠進(jìn)入開(kāi)放臂的次數(shù)(OE)、開(kāi)放臂停留的時(shí)間(OT)、進(jìn)入開(kāi)放臂的次數(shù)百分比(OE%)和進(jìn)入開(kāi)放臂的時(shí)間百分比(OT%)均顯著降低(p0.01);模型組大鼠運(yùn)動(dòng)的總路程、中央活動(dòng)路程百分比和中央活動(dòng)時(shí)間百分比均顯著降低(p0.01,p0.05)。(3)在基礎(chǔ)條件下,兩組大鼠分泌的促腎上腺皮質(zhì)激素和皮質(zhì)酮無(wú)明顯差異(p0.05);而在應(yīng)激條件下,模型組大鼠分泌的兩種激素增加顯著高于對(duì)照組(p0.01)。(4)與對(duì)照組相比,模型組MR m RNA在大鼠海馬組織中表達(dá)無(wú)明顯差異(p0.05);而模型組大鼠GR在海馬、下丘腦、垂體和腎上腺組織中的m RNA表達(dá)水平均顯著降低(p0.01,p0.05)。(5)與對(duì)照組比較,模型組MR蛋白表達(dá)在大鼠海馬組織中無(wú)明顯差異(p0.05);GR蛋白表達(dá)在大鼠海馬、下丘腦和腎上腺組織中均顯著降低(p0.01,p0.05),而在垂體中表達(dá)無(wú)差異(p0.05)。結(jié)論:在應(yīng)激條件下,焦慮模型大鼠HPA軸呈高反應(yīng)性與GR介導(dǎo)的軸負(fù)反饋調(diào)節(jié)作用改變相關(guān)。
[Abstract]:Objective: To study the anxiety under stress in rat model of hypothalamic pituitary adrenal axis (hypothalamus-pituitary-adrenal, HPA) showed high reactivity with the shaft itself or hyperfunction associated with corticosteroid receptor changes. Methods: healthy male SD rats were randomly divided into two groups, the control group (n=20) and model group (n=26) to establish a rat model of anxiety. The emotional stress method is unpredictable, the control group was given normal diet. All rats were weighed every week in weight, and after the end of the modeling by behavioral experiments testing the success of modeling. The elevated plus maze and open field as a source of stress stimulation in rats, the determination of the two groups of rats in the serum of rats with adrenocorticotropic hormone (ACTH) and corticosterone (CORT) levels. Detected by real-time fluorescence quantitative PCR in the hippocampus of rats in the two groups mineralocorticoid receptor (mineralocorticoid steroid, receptor, MR) and sugar Glucocorticoid receptor (glucocorticoid steroid, receptor, GR) m RNA level and GR expression level of M RNA in hypothalamus, pituitary and adrenal tissues. Western blot (Western blot) detection in the hippocampus of rats in the two groups of MR and GR protein level and GR expression level in hypothalamus, pituitary and adrenal tissue protein results: (1) compared with the control group, the anxiety model rats weight growth rate decreased significantly (P0.01). (2) the elevated plus maze and open field test results show that, compared with the control group, the number of rats in the open arm (OE), open arm time (OT). The percentage of times in the open arm (OE%) and in the open arm time percentage (OT%) were significantly decreased (P0.01); the total distance of rats in the model group movement, the percentage of the central distance percentage and central activity time were significantly decreased (P0.01, P0.05). (3) based on conditions Next, two groups of adrenocorticotropic hormone and corticosterone secretion in rats had no significant difference (P0.05); while under stress condition, two kinds of models of hormone secretion in rats group increased significantly higher than the control group (P0.01). (4) compared with the control group, model group, MR m RNA in rat hippocampal tissue there was no difference between the expressions (P0.05); while the rats in the model group GR in hippocampus, hypothalamus and pituitary, the expression level of M RNA in adrenal glands were significantly decreased (P0.01, P0.05). (5) compared with the control group, model group, the expression of MR protein in hippocampus of rats had no obvious difference (P0.05); the expression of GR protein in rat hippocampus, hypothalamus and adrenal tissues were significantly decreased (P0.01, P0.05), and no difference in pituitary expression (P0.05). Conclusion: under stress, anxiety model rat HPA Axis axis showed high reactivity and negative feedback regulation mediated by GR is related to the change of.
