【摘要】：目的(1)檢驗5-羥色胺轉(zhuǎn)運體基因啟動子區(qū)域連鎖多態(tài)性(5-HTTLPR)對青少年抑郁癥狀的影響；(2)檢驗5-HTTLPR與應(yīng)激性生活事件的交互效應(yīng)對抑郁癥狀的預(yù)測作用。 方法隨機抽取湖南省長沙市某所高中的252名高一年級學生,并進行為期2年共9次的追蹤研究。初次測評時,分別用流調(diào)中心用抑郁量表(Center for Epidemiological Studies Depression Scale, CES-D)和青少年生活事件問卷(Adolescent Life Events Questionnaire, ALEQ)評估被試的抑郁癥狀和生活事件,并抽取5mL靜脈血以提取DNA和鑒定5-HTTLPR基因型。初測后的2年中,每隔3個月對所有被試的抑郁癥狀和生活事件進行追蹤評估,共8次追蹤。運用多層線性模型(Hierarchical Linear Model, HLM)對收集的數(shù)據(jù)進行統(tǒng)計分析。 結(jié)果多層線性模型分析顯示：(1)生活事件對男性和女性的抑郁癥狀均有顯著預(yù)測作用(男性：β=0.14,p0.0001；女性：β=0.13,p0.0001);(2)5-HTTLPR對男性和女性的抑郁癥狀的預(yù)測作用均無主效應(yīng)(男性：β=0.21,p0.05；女性：β=1.16,p0.05)；(3)女性樣本中,5-HTTLPR與生活事件交互作用顯著(β=0.08,p0.0001),攜帶S等位基因型的個體在應(yīng)激性生活事件后會表現(xiàn)出更高的抑郁癥狀；男性樣本中,無顯著交互作用(β=-0.03,p0.05)。 結(jié)論5-HTTLPR對青少年抑郁癥狀無預(yù)測作用；5-HTTLPR與應(yīng)激的交互作用能夠預(yù)測女性青少年的抑郁癥狀,而對男性青少年抑郁癥狀沒有預(yù)測作用。
[Abstract]:Objective to test the effect of 5-HTTLPRs on depression symptoms in adolescents by using 5-HTTLPR) to test the predictive effect of 5-HTTLPR and stress life events on depression symptoms. Methods A total of 252 freshmen from a high school in Changsha, Hunan Province, were randomly selected and followed up for 2 years and 9 times. Depression symptoms and life events were assessed with the Center for Epidemiological Studies Depression scale (CES-D) and Adolescent Life Events questionnaire (ALEQs), and 5 mL venous blood was extracted to extract DNA and identify 5-HTTLPR genotypes. The depressive symptoms and life events of all subjects were tracked and evaluated every 3 months, and the collected data were statistically analyzed by using the hierarchical Linear Model (HLMModel). Results the multilevel linear model analysis showed that life events had significant predictive effects on both male and female depressive symptoms (male: 尾 = 0.14, p 0.0001; female: 尾 = 0.13, p 0.0001). There was no significant predictive effect of 5-HTTLPR on depressive symptoms in both males and females (male: 尾 0.21, p0.05; female: 尾: 尾 0.21, p0.05; male: 尾 0.21, p0.05; female: 尾: 尾 0.21, p0.0001; male: 尾 0.21, p0.05; male: 尾 0.21, p0.0001; Sex: in female samples, the interaction between 5-HTTLPR and life events was significant (尾 0.08% p0.0001). Individuals with S allele showed higher depressive symptoms after the stress sexual events. There was no significant interaction in male samples (尾 -0.03 P 0.05). Conclusion there is no predictive effect of 5-HTTLPR on depression symptoms in adolescents. The interaction between 5-HTTLPR and stress can predict depression symptoms in female adolescents, but not in male adolescents.
