鷹嘴豆芽素A對(duì)過氧化氫誘導(dǎo)大鼠原代海馬神經(jīng)元損傷的作用及抗凋亡機(jī)制

【摘要】:目的探討植物雌激素鷹嘴豆芽素A對(duì)H_2O_2誘導(dǎo)大鼠原代海馬神經(jīng)元細(xì)胞凋亡的保護(hù)作用。方法取新生24 h內(nèi)的海馬神經(jīng)元細(xì)胞,培養(yǎng)10 d左右,鑒定神經(jīng)細(xì)胞元純度。用雌激素和低、中、高劑量的鷹嘴豆芽素A處理細(xì)胞24 h,除正常組外,其余組給予H_2O_2作用12 h,CCK-8法測(cè)定細(xì)胞活力,用流式細(xì)胞儀測(cè)定各組細(xì)胞凋亡率。Western blot測(cè)定Bcl-2、Bax、caspase-3蛋白表達(dá)水平。結(jié)果用神經(jīng)元特異性抗體微管相關(guān)蛋白2熒光檢測(cè)海馬神經(jīng)元純度達(dá)98%以上,鷹嘴豆芽素A預(yù)處理24 h能顯著改善H_2O_2作用12 h后的海馬神經(jīng)元活力。用流式細(xì)胞儀觀察H_2O_2損傷后,凋亡細(xì)胞數(shù)和壞死細(xì)胞數(shù)明顯增加,給予鷹嘴豆芽素A預(yù)處理后,可明顯減少H_2O_2引起的凋亡和壞死細(xì)胞數(shù)。鷹嘴豆芽素A各劑量組劑量依賴性地提高模型細(xì)胞Bcl-2蛋白表達(dá),降低Bax、caspase-3蛋白表達(dá)。結(jié)論?dān)椬於寡克谹對(duì)H_2O_2誘導(dǎo)大鼠原代海馬神經(jīng)元損傷的保護(hù)作用可能與其抑制細(xì)胞凋亡有關(guān),鷹嘴豆芽素A抗神經(jīng)元凋亡作用的機(jī)制可能與其上調(diào)Bcl-2蛋白表達(dá)和下調(diào)Bax、caspase-3蛋白表達(dá)有關(guān)。
[Abstract]:Objective to investigate the protective effect of phytoestrogenin A on apoptosis of primary hippocampal neurons induced by H _ 2O _ 2 in rats. In addition to the normal group, the other groups were treated with H _ 2O _ 2 for 12 h to determine the cell viability by CCK-8 method. Apoptosis rate was measured by flow cytometry. Western blot was used to detect the expression of Bcl-2Baxax-caspase-3 protein. Results the purity of hippocampal neurons was more than 98% by fluorescence detection of neuron specific antibody microtubule-associated protein 2. The hippocampal neuronal activity of H2O2 was significantly improved after H2O2 was pretreated for 24 h. The number of apoptotic cells and necrotic cells increased significantly after H _ 2O _ 2 injury was observed by flow cytometry, and the number of apoptotic cells and necrotic cells were observed by flow cytometry. The number of apoptotic and necrotic cells induced by H _ 2O _ 2 was significantly reduced. The expression of Bcl-2 protein in model cells was increased in a dose-dependent manner. Conclusion the protective effect of chickpein A on the injury of primary hippocampal neurons induced by S _ 2O _ 2 in rats may be related to its inhibition of apoptosis. The mechanism of antiapoptotic effect of chickpein A may be related to its up-regulation of Bcl-2 protein expression and down-regulation of Bax-caspase-3 protein expression.
