GABA_A受體激動劑對AD模型配體門控氯通道的作用

【摘要】:目的觀察并分析GABA_A受體激動劑蠅蕈醇對GABA激活的配體門控氯通道的作用,為尋找新的抗阿爾茨海默病(AD)藥物提供實驗依據(jù)。方法 Aβ_(1-42)誘導(dǎo)海馬神經(jīng)細胞作為AD細胞模型;通過膜片鉗(Patch clamp)技術(shù)在大鼠海馬神經(jīng)元上記錄配體門控氯通道瞬時外向電流,采用非特異性氯通道阻斷劑(NFA)和無GABA的細胞外液確定該電流成份為瞬時外向氯電流;通過膜片鉗技術(shù)觀察GABA_A受體激動劑蠅蕈醇對于在大鼠海馬神經(jīng)元上記錄瞬時外向電流的作用;向AD細胞模型中加入GABA_A受體激動劑,采用MTT觀察各組細胞活力。結(jié)果加入GABA_A受體激動劑蠅蕈醇后GABA激活氯電流強度增加;加入濃度為1 mmol/L蠅蕈醇的AD模型細胞組存活率為(91.32±3.73)%,略高于未加入蠅蕈醇的模型細胞組,兩組對比P0.05。結(jié)論 GABA_A受體激動劑蠅蕈醇對于AD細胞模型具有保護作用。
[Abstract]:Objective to observe and analyze the effect of muscarinol, a GABA_A receptor agonist, on ligand gated chloride channels activated by GABA, and to provide experimental evidence for finding new drugs against Alzheimer's disease. Methods A 尾 -Stack 1-42) induced hippocampal neurons as AD cell model. The transient outward currents of ligand gated chloride channels were recorded in rat hippocampal neurons by patch clamp clamp technique. The transient outward chloride currents were determined by non-specific chloride channel blockers (NFAs) and extracellular fluids without GABA. The effects of muscarinol, a GABA_A receptor agonist, on recording transient outward currents in rat hippocampal neurons were observed by patch clamp technique, and GABA_A receptor agonists were added to AD cell model. Results after adding GABA_A receptor agonist muscarinol, the activity of GABA increased, and the survival rate of AD model cells with 1 mmol/L muscarinol was 91.32 鹵3.73, which was slightly higher than that of model cells without muscarinol. Conclusion Muscarinol, a GABA_A receptor agonist, has protective effect on AD cell model.
