焦慮性抑郁模型大鼠腦區(qū)單胺遞質(zhì)含量與神經(jīng)因子表達(dá)的變化

【摘要】:目的研究焦慮性抑郁模型大鼠海馬、杏仁核、前額葉皮質(zhì)內(nèi)單胺遞質(zhì)的含量變化及腦內(nèi)神經(jīng)營養(yǎng)因子的表達(dá)趨勢,探討其可能的發(fā)病機(jī)制。方法 60只SD大鼠隨機(jī)分為正常對照組、溶媒對照組、焦慮模型組、抑郁模型組、焦慮性抑郁模型組,每組12只。采用慢性束縛應(yīng)激聯(lián)合皮質(zhì)酮注射的方法建立焦慮性抑郁大鼠模型,造模時間為21 d,造模結(jié)束后采用高架十字迷宮測試,曠場實驗,強(qiáng)迫游泳實驗評價大鼠的焦慮和抑郁樣行為,HPLC-ECD法檢測大鼠海馬、杏仁核、前額葉皮質(zhì)的單胺遞質(zhì)5-HT、NE、DA含量,蛋白印跡法檢測大鼠各腦區(qū)神經(jīng)營養(yǎng)因子BDNF、NT-3的含量。結(jié)果焦慮性抑郁模型組大鼠在進(jìn)入開臂的時間、次數(shù)、曠場中自主活動次數(shù)均與焦慮組相當(dāng),與對照組及抑郁組比較差異有顯著性(P0.01或P0.05),在強(qiáng)迫游泳中的不動時間顯著增加,與對照組及焦慮組對比差異有顯著性(P0.01);同時,與對照組比較,焦慮性抑郁模型組大鼠海馬5-HT、杏仁核及前額葉皮質(zhì)區(qū)的5-HT和NE含量均顯著下降(P0.01或P0.05);此外,與對照組比較,焦慮性抑郁模型組大鼠各腦區(qū)BDNF、NT-3含量顯著下降(P0.01或P0.05),同時與焦慮組比較,BDNF含量顯著下降(P0.05)。結(jié)論焦慮性抑郁模型組大鼠具有顯著的焦慮及抑郁樣行為,其發(fā)病機(jī)制可能與腦內(nèi)海馬、杏仁核、前額葉皮質(zhì)區(qū)域的單胺遞質(zhì)含量降低及神經(jīng)營養(yǎng)因子BDNF、NT-3表達(dá)下調(diào)有關(guān)。
[Abstract]:Objective to study the rat hippocampus, amygdala anxiety depression model, expression trend and content changes of prefrontal cortex monoamine neurotransmitters in neurotrophic factors, to explore its possible pathogenesis. Methods: 60 SD rats were randomly divided into normal control group, control group, anxiety group, depression group, anxiety depression model group, 12 rats in each group. Using the method of chronic restraint stress combined with corticosterone injection of rat models of anxiety and depression, the modeling time is 21 d, after the modeling using the elevated plus maze test, open field test, forced swimming test evaluation of rat anxiety and depression like behavior, HPLC-ECD assay rat hippocampus, amygdala, prefrontal cortex monoamine neurotransmitters 5-HT, NE, DA content, Western blot detection of rat brain neurotrophic factor BDNF, the content of NT-3. Results the anxiety depression model rats in the open arm Time, the number of open field in the number of spontaneous activity and anxiety group, there was significant difference compared with the control group and depression group (P0.01 or P0.05), a significant increase in immobility time of forced swimming, there was significant difference between group and anxiety group compared with the control (P0.01); at the same time, with the control group comparison of anxiety in 5-HT model rats of depression, 5-HT and NE in the amygdala and prefrontal cortex were significantly decreased (P0.01 or P0.05); in addition, compared with the control group, cerebral BDNF anxiety and depression model rats, NT-3 content decreased significantly (P0.01 or P0.05), and compared with anxiety group, BDNF content decreased significantly (P0.05). Significant anxiety and depression like behavior has conclusion anxiety and depression model rats, its mechanism may be related to the brain hippocampus, amygdala, prefrontal cortex monoamine neurotransmitter contents decreased and neurotrophic factor BDNF, NT-3 expression It's related to the tune.
