丙酮酸鹽對(duì)阿爾茨海默病大鼠學(xué)習(xí)記憶的影響

【摘要】:目的研究丙酮酸鹽對(duì)阿爾茨海默病(Alzheimer’s disease,AD)模型大鼠學(xué)習(xí)記憶的影響并探討其與海馬神經(jīng)細(xì)胞死亡的關(guān)系。方法將36只wistar大鼠隨機(jī)分為空白對(duì)照組、AD模型組和丙酮酸鹽治療組,根據(jù)實(shí)驗(yàn)要求給藥處理6周,同時(shí)觀察各組大鼠一般情況,Morris水迷宮實(shí)驗(yàn)檢測(cè)大鼠學(xué)習(xí)記憶能力,蘇木精-伊紅(hematoxylin and eosin,HE)染色檢測(cè)海馬神經(jīng)細(xì)胞死亡,利用2′,7′-dichlorodihydrofluorescein diacetate(H2DCF-DA)來檢測(cè)海馬CA1區(qū)內(nèi)活性氧(reactive oxygen species,ROS)聚集情況。結(jié)果丙酮酸鹽治療組與AD模型組比較,一般情況均明顯改善,逃避潛伏期均明顯縮短(41.37±6.24,31.67±7.26,24.26±7.16,20.45±5.72,19.36±5.89,均P0.05);原平臺(tái)象限活動(dòng)時(shí)間明顯增加(50.36±7.46 s,P0.05),穿越原平臺(tái)次數(shù)明顯增多(3.98±1.23次/2 min,P0.05),海馬CA1區(qū)錐體神經(jīng)細(xì)胞死亡顯著減少(P0.05),海馬神經(jīng)細(xì)胞內(nèi)ROS的水平也顯著降低(P0.05)。結(jié)論丙酮酸鹽可能通過降低海馬組織內(nèi)ROS的產(chǎn)生,減少海馬神經(jīng)細(xì)胞死亡,從而提高AD模型大鼠的學(xué)習(xí)記憶水平。
[Abstract]:Objective to study the effect of pyruvate on learning and memory in rats with Alzheimer's disease (AD) and to explore the relationship between pyruvate and hippocampal nerve cell death. Methods 36 wistar rats were randomly divided into control group and pyruvate treatment group. According to the experimental requirements, the rats were treated for 6 weeks, and their learning and memory abilities were measured by Morris water maze test and hematoxylin and eosin HEH staining to detect the death of hippocampal nerve cells. The aggregation of reactive oxygen speciess-ROSs in the hippocampal CA1 was detected by using 2-dichlorodihydrofluorescein diacetate H2DCF-DA.Results the general situation of pyruvate treatment group was significantly improved compared with that of AD model group. The latency of escape was significantly shortened by 41.37 鹵6.24s31.67 鹵7.26tl 24.26 鹵7.162n 20.45 鹵5.72n 19.36 鹵5.89m, and the time of activity in the original platform quadrant was significantly increased by 50.36 鹵7.46sm P0.05n, and the number of times of crossing the original platform was increased by 3.98 鹵1.23 times per second minute P0.05. the death of pyramidal neurons in hippocampal CA1 region significantly decreased P0.05a, and the ROS of hippocampal neurons in the hippocampal neurons decreased significantly. Conclusion Pyruvate may decrease the production of ROS in hippocampal tissue. Decrease the death of hippocampal nerve cells and improve the learning and memory level of AD model rats.
