【摘要】：目的用免疫學(xué)和分子生物學(xué)方法調(diào)查貴州省精神分裂癥患者弓形蟲的感染狀況，并用多重巢式-聚合酶鏈反應(yīng)-限制性片段長(zhǎng)度多態(tài)性(Mn-PCR-RFLP)方法進(jìn)行弓形蟲基因分型，探討精神分裂癥與弓形蟲感染的關(guān)系以及貴州省弓形蟲感染的分子流行病學(xué)特點(diǎn)。方法采集112份精神分裂癥患者（疾病組）和101份正常人群（對(duì)照組）外周血標(biāo)本。采用ELISA法檢測(cè)外周血血漿中弓形蟲循環(huán)抗原(CAg)及抗弓形蟲特異性IgM、IgG抗體。應(yīng)用PCR技術(shù)擴(kuò)增外周血弓形蟲的特異性基因片段(529bp)，檢測(cè)弓形蟲核酸。對(duì)于弓形蟲核酸檢測(cè)陽(yáng)性的標(biāo)本用Mn-PCR-RFLP法分析弓形蟲c22-8， c29-2，， L358，PK1，SAG2(5`-SAG2，3`-SAG2，alt.SAG2)，BTUB，GRA6，SAG3，Apico，SAG1共10個(gè)不同基因位點(diǎn)，鑒定弓形蟲的基因型，分析貴州省精神分裂癥患者弓形蟲感染的分子流行病學(xué)特點(diǎn)。結(jié)果112份精神分裂癥患者標(biāo)本中檢測(cè)到弓形蟲特異性IgG抗體陽(yáng)性18例， CAg陽(yáng)性1例，弓形蟲核酸陽(yáng)性6例，101份正常人群標(biāo)本中檢測(cè)到弓形蟲特異性IgG抗體陽(yáng)性7例， CAg陽(yáng)性1例。精神分裂癥患者弓形蟲感染率為22.3%，與對(duì)照組（7.9%）比較，差異具有統(tǒng)計(jì)學(xué)意義(P=0.0042)。測(cè)得的OR值為3.391，95%CI=1.420-8.097。Mn-PCR-RFLP鑒定6份標(biāo)本中弓形蟲的基因型一致,屬于非典型蟲株ToxoDB#9。結(jié)論精神分裂患者弓形蟲感染率高于正常人群，提示精神分裂癥的發(fā)生可能與弓形蟲感染有關(guān)。貴州省精神分裂癥患者感染的弓形蟲屬于不典型基因型Chinese1。
[Abstract]:Objective to investigate the infection of Toxoplasma gondii in schizophrenic patients in Guizhou Province by immunological and molecular biological methods. The genotyping of Toxoplasma gondii was carried out by multiplex nested polymerase chain reaction-restriction fragment length polymorphism (Mn-PCR-RFLP). To explore the relationship between schizophrenia and Toxoplasma gondii infection and the molecular epidemiological characteristics of toxoplasmosis infection in Guizhou Province. Methods 112 schizophrenic patients (disease group) and 101 normal persons (control group) were collected. The circulating antigen of Toxoplasma gondii (Toxoplasma gondii) and anti-Toxoplasma gondii (Toxoplasma gondii) specific IgM were detected by ELISA method. IgG antibody. The specific gene fragment of Toxoplasma gondii was amplified by PCR. Toxoplasma gondii nucleic acid was detected. Toxoplasma gondii c22-8, c29-2, L358PK1 were analyzed by Mn-PCR-RFLP method. SAG2(5 '-SAG2O3`-SAG2Alt.SAG2ANG2, BTUBMAGRA6, SAG3, Apicotrol SAG1 have 10 different gene loci. Identification of Toxoplasma gondii genotypes. The molecular epidemiologic characteristics of Toxoplasma gondii infection in schizophrenic patients in Guizhou Province were analyzed. Results 18 cases of Toxoplasma gondii specific IgG antibody were detected in 112 samples of schizophrenic patients. CAg was positive in 1 case, Toxoplasma nucleic acid positive in 6 cases and Toxoplasma gondii specific IgG antibody in 7 cases. The infection rate of Toxoplasma gondii in schizophrenic patients was 22. 3%, which was compared with that in control group (7. 9%). The difference was statistically significant. The OR was 3.391. The genotypes of Toxoplasma gondii were identical in 6 samples identified by 95CII 1.420-8.097.Mn-PCR-RFLP. Conclusion Toxoplasma gondii infection rate in schizophrenic patients is higher than that in normal controls. The results suggest that the occurrence of schizophrenia may be related to Toxoplasma gondii infection. Toxoplasma gondii infected by schizophrenic patients in Guizhou Province belongs to the atypical genotype Chinese1.
