成熟中性粒細(xì)胞中淀粉樣變纖維成分的檢測(cè)

【摘要】:目的:先前報(bào)道紅細(xì)胞中存在淀粉樣纖維,本文進(jìn)一步探討成熟中性粒細(xì)胞是否存在淀粉樣變纖維,為解析阿爾茨海默病(AD)淀粉樣變形成分子機(jī)制提供科學(xué)依據(jù)。方法:收集AD患者以及對(duì)照人群各30例,利用希森美康X5-500i血液分析儀測(cè)定血液白細(xì)胞、紅細(xì)胞、血紅蛋白水平以及中性粒細(xì)胞絕對(duì)值等4項(xiàng)指標(biāo),采集靜脈EDTA-K_2抗凝血分離中性粒細(xì)胞,分別測(cè)定中性粒細(xì)胞與淀粉樣變特異探針之間親和性,血漿部分用硫磺素T(ThT)檢測(cè)淀粉樣變熒光強(qiáng)度。結(jié)果:親和試驗(yàn)結(jié)果顯示淀粉樣變熒光探針(ThT)能夠與組織淀粉樣變纖維特異性結(jié)合,中性粒細(xì)胞中淀粉樣變纖維染色也呈現(xiàn)陽(yáng)性反應(yīng)。AD組與健康對(duì)照組間血細(xì)胞白細(xì)胞、紅細(xì)胞、血紅蛋白水平以及中性粒細(xì)胞絕對(duì)值比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),但是血漿淀粉樣變熒光強(qiáng)度顯著高于對(duì)照組(P0.05)。結(jié)論:報(bào)道成熟中性粒細(xì)胞發(fā)現(xiàn)淀粉樣變纖維,且AD患者血漿淀粉樣變纖維水平顯著增高。
[Abstract]:Objective: previous reports exist of amyloid fibrils in red blood cells, this paper further explores the existence of mature neutrophils to amyloid fibrils, for the analysis of Alzheimer's disease (AD) amyloid formation molecular mechanism to provide scientific basis. Methods: collect AD patients and control group 30 cases, determination of white blood cell, using Sysmex X5-500i of RBC, hemoglobin level and 4 indexes of neutrophils, collecting venous EDTA-K_2 anticoagulant isolated neutrophils, neutrophils were determined with amyloid specific probe affinity between plasma respectively, with Thioflavin T (ThT) to detect amyloid fluorescence intensity. Results: affinity test the results showed that amyloid fluorescence probe (ThT) can be combined with tissue amyloid fibers, neutrophil amyloid fiber staining also showed positive reaction in.AD group With the healthy control group between blood cells and white blood cells, red blood cells, hemoglobin level and absolute neutrophil count had no statistically significant difference (P0.05), but serum amyloid fluorescence intensity was significantly higher than the control group (P0.05). Conclusion: reports of mature neutrophils are amyloid fibrils, and plasma AD of patients with starch like fiber level increased significantly.
