旋毛蟲(chóng)成蟲(chóng)排泄分泌蛋白抗CLP誘導(dǎo)的小鼠膿毒癥的觀察

【摘要】:目的探討旋毛蟲(chóng)成蟲(chóng)排泄分泌蛋白(AES)對(duì)盲腸結(jié)扎穿孔術(shù)(CLP)誘導(dǎo)的小鼠膿毒癥的抑制作用。方法將48只BALB/c小鼠隨機(jī)分為3組,即假手術(shù)對(duì)照組(PBS+sham組,A組)、膿毒癥組(PBS+CLP組,B組)、AES蛋白治療組(AES+CLP組,C組)。A組和B組于CLP術(shù)后經(jīng)腹腔注射200μl PBS,C組術(shù)后經(jīng)腹腔注射含25μg AES蛋白的等量PBS。每組隨機(jī)抽取8只,建模后觀察各組小鼠的一般情況及96 h生存率;每組其余8只小鼠于建模后12 h取肝、肺、腎,HE染色觀察病理改變;以酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測(cè)小鼠血清中TNF-α、IL-1β、IL-6、IL-10及TGF-β細(xì)胞因子水平的變化。結(jié)果 3組小鼠96 h生存率差異有統(tǒng)計(jì)學(xué)意義(χ~2=21.16,P0.05);與A組(100%)相比,B組小鼠生存率(0)降低(P0.05),且肺、肝及腎臟損傷明顯加重;經(jīng)AES治療后,C組存活率(75%)明顯增加(P0.05),小鼠肺臟、肝臟及腎臟病理?yè)p傷亦顯著緩解。3組小鼠血清中的促炎因子TNF-α(F=27.11,P0.05)、IL-1β(F=18.75,P0.05)及IL-6(F=100.93,P0.05)水平差異均有統(tǒng)計(jì)學(xué)意義;B組小鼠的促炎因子水平均高于A組(P均0.05);經(jīng)AES治療后,C組小鼠上述3種促炎因子水平較B組均顯著下降(P均0.05)。3組小鼠血清中的調(diào)節(jié)性細(xì)胞因子IL-10(F=10.88,P0.05)及TGF-β(F=11.37,P0.05)水平差異亦有統(tǒng)計(jì)學(xué)意義;經(jīng)AES治療后,C組小鼠IL-10及TGF-β分泌水平較B組亦明顯增加(P0.05)。結(jié)論旋毛蟲(chóng)AES對(duì)CLP誘導(dǎo)的BALB/c小鼠膿毒癥有顯著的緩解作用。
[Abstract]:Objective to investigate the inhibitory effect of excretion and secretory protein (AES) of Trichinella spiralis on sepsis induced by cecal ligation and perforation in mice. Methods 48 BALB/c mice were randomly divided into three groups: pseudo-operation control group (PBS sham group (group A), sepsis group (PBS CLP group, group B), AES protein treatment group (AES CLP group, group C). A group and group B after intraabdominal injection of 200 渭 l PBS,C group. The same amount of PBS. containing 25 渭 g AES protein was injected intraperitoneally after CLP operation. Eight mice in each group were randomly selected to observe the general situation and 96 h survival rate of mice in each group, and the pathological changes of liver, lung, kidney and HE were observed at 12 h after modeling, and the levels of TNF- 偽, IL-1 尾, IL-6,IL-10 and TGF- 尾 cytokines in serum were detected by enzyme linked immunosorbent assay (ELISA). Results the 96 h survival rate of mice in group B was significantly lower than that in group A (P < 0.05), and the survival rate in group B was significantly lower than that in group A (P 0.05), and the injury of lung, liver and kidney was significantly worse in group B than that in group A (P < 0.05), and the survival rate in group B was significantly lower than that in group A (P 0.05).
