HMGB1與自噬信號Beclin1在急性胰腺炎中的相關(guān)性研究

【摘要】:目的:研究HMGB1在自噬信號分子Beclin1過度激活的急性胰腺炎中的表達意義,為臨床治療和探討急性胰腺炎的發(fā)生發(fā)展機制提供新的理論依據(jù)。 方法:(1)取44只SD大鼠(350±30g)隨機分為假手術(shù)組(SO)(n=12)和實驗組(ANP)(n=32),實驗組以5%牛黃膽酸鈉胰膽管逆行注射制作急性胰腺炎大鼠模型,分別給予造模3h、6h、12h、24h后進行麻醉后心臟放血處死。(2)收集血液,離心取上清,采用Elisa方法檢測外周血液中淀粉酶及炎癥因子HMGB1、TNF-α、IL-1和IL-6的含量。(3)同時取胰腺組織做HE染色進行病理觀察,并采用免疫組化實驗檢測胰腺組織中HMGB1及Beclinl的表達,并利用軟件將組化結(jié)果進行定量分析。 結(jié)果:(1)急性胰腺炎造模成功,模型組血清淀粉酶明顯高于假手術(shù)組(P0.05);外周血檢測,早期炎癥因子IL-1、TNF-α、IL-6在造模3h后表達明顯升高,6h水平達峰值,且高于假手術(shù)組(P0.05),12h后出現(xiàn)表達的下調(diào),24h后炎癥因子表達低于12h水平(P0.05);HMGB1在3h無明顯變化(P0.05),6h時明顯增高,后繼續(xù)維持較高水平(P0.05)。(2)在急性胰腺炎組織免疫組化中,HMGB1在急性壞死性胰腺炎(ANP組)3h、6h組無明顯變化(P0.05),在ANP組12h后明顯升高,且在24h后持續(xù)升高,具有統(tǒng)計學(xué)意義(P0.05);Beclinl隨著炎癥的加重與炎癥反應(yīng)時間的推移,表達持續(xù)增高,具有統(tǒng)計學(xué)意義(P0.05)。 結(jié)論: (1)作為晚期炎癥因子的HMGB1在急性胰腺炎中表達明顯晚于早期炎癥因子IL-1、TNF-α、IL-6,可能在維持和促進急性胰腺炎的發(fā)生發(fā)展存在關(guān)鍵作用。 (2)HMGB1可能對自噬信號Beclin1在急性胰腺炎中的表達起到維持和促進作用,從而促進急性胰腺的病程進展。
