人膝關(guān)節(jié)滑膜細(xì)胞的增殖、分泌和趨化的體外研究

【摘要】:第一部分人膝關(guān)節(jié)滑膜細(xì)胞的體外培養(yǎng) 目的:獲得人滑膜B型細(xì)胞,進(jìn)行后續(xù)研究。方法:提取滑膜組織,消化,培養(yǎng),傳代,鑒定。結(jié)果:獲得人滑膜B型細(xì)胞。結(jié)論:本方法可以獲得人滑膜B型細(xì)胞,滿足實(shí)驗(yàn)的需求。 第二部分tTG. β-NGF及bFGF對(duì)人膝關(guān)節(jié)滑膜細(xì)胞的作用 目的:研究tTG、βNGF及bFGF對(duì)人膝關(guān)節(jié)滑膜B型細(xì)胞的作用。方法:將第一部分培養(yǎng)好的滑膜細(xì)胞鋪板,加入不同濃度的tTG、β-NGF及bFGF,抽取上清,測(cè)Ⅰ型膠原蛋白的含量,加入MTT,測(cè)細(xì)胞增殖,利用Transwel l測(cè)趨化作用。結(jié)果:tTG在20ng/ml之前,對(duì)滑膜B型細(xì)胞有促進(jìn)增殖、分泌作用,之后,隨著濃度的增加,抑制細(xì)胞增殖和分泌Ⅰ型膠原蛋白,tTG對(duì)滑膜B型細(xì)胞無(wú)趨化作用。β-NGF對(duì)滑膜B型細(xì)胞有抑制增殖和分泌Ⅰ型膠原蛋白作用,β-NGF對(duì)滑膜B型細(xì)胞無(wú)趨化作用。bFGF對(duì)滑膜B型細(xì)胞有促進(jìn)增殖、分泌作用,bFGF對(duì)滑膜B型細(xì)胞無(wú)趨化作用。結(jié)論:在滑膜B型細(xì)胞增殖和分泌Ⅰ型膠原蛋白方面,tTG有雙重作用,β-NGF有抑制作用,bFGF有促進(jìn)作用;在滑膜B型細(xì)胞趨化方面,三種均無(wú)影響。
