【摘要】：第一部分人膝關節(jié)滑膜細胞的體外培養(yǎng) 目的：獲得人滑膜B型細胞,進行后續(xù)研究。方法：提取滑膜組織,消化,培養(yǎng),傳代,鑒定。結果：獲得人滑膜B型細胞。結論：本方法可以獲得人滑膜B型細胞,滿足實驗的需求。 第二部分tTG. β-NGF及bFGF對人膝關節(jié)滑膜細胞的作用 目的：研究tTG、βNGF及bFGF對人膝關節(jié)滑膜B型細胞的作用。方法：將第一部分培養(yǎng)好的滑膜細胞鋪板,加入不同濃度的tTG、β-NGF及bFGF,抽取上清,測Ⅰ型膠原蛋白的含量,加入MTT,測細胞增殖,利用Transwel l測趨化作用。結果：tTG在20ng/ml之前,對滑膜B型細胞有促進增殖、分泌作用,之后,隨著濃度的增加,抑制細胞增殖和分泌Ⅰ型膠原蛋白,tTG對滑膜B型細胞無趨化作用。β-NGF對滑膜B型細胞有抑制增殖和分泌Ⅰ型膠原蛋白作用,β-NGF對滑膜B型細胞無趨化作用。bFGF對滑膜B型細胞有促進增殖、分泌作用,bFGF對滑膜B型細胞無趨化作用。結論：在滑膜B型細胞增殖和分泌Ⅰ型膠原蛋白方面,tTG有雙重作用,β-NGF有抑制作用,bFGF有促進作用；在滑膜B型細胞趨化方面,三種均無影響。
