鮑曼不動(dòng)桿菌膿毒癥大鼠體內(nèi)TCTP表達(dá)情況及血必凈對(duì)其表達(dá)的影響

【摘要】:目的:通過研究鮑曼不動(dòng)桿菌膿毒癥大鼠中性粒細(xì)胞和肝臟組織中翻譯控制腫瘤蛋白(Thetranslationallycontrolledtumorprotein,TCTP)在不同時(shí)間點(diǎn)的表達(dá)情況,并以血必凈進(jìn)行干預(yù)治療,檢測炎癥因子IL-6、IL-10及肝臟組織病理變化,探討TCTP與鮑曼不動(dòng)桿菌膿毒癥之間的關(guān)系,及血必凈對(duì)其表達(dá)的影響。 方法:1、42只清潔級(jí)成年雄性wistar大鼠隨機(jī)分成7組:正常對(duì)照組(N),鮑曼不動(dòng)桿菌膿毒癥6h、12h、24h組(B6h、B12h、B24h),血必凈治療6h、12h、24h組(X6h、X12h、X24h)。其中鮑曼不動(dòng)桿菌膿毒癥各組及血必凈治療各組大鼠分別腹腔注射鮑曼不動(dòng)桿菌懸液,正常對(duì)照組大鼠腹腔注射等量生理鹽水;血必凈治療各組大鼠尾靜脈注射血必凈,余各組大鼠尾靜脈注射等量生理鹽水。2、正常組、不同時(shí)間點(diǎn)鮑曼不動(dòng)桿菌膿毒癥組及血必凈治療組大鼠頸動(dòng)脈采血后取肝臟,用westernblot檢測中性粒細(xì)胞內(nèi)TCTP的表達(dá)變化,ELISA檢測血漿中炎癥因子IL-6、IL-10的變化,,用westernblot及免疫組化檢測肝臟組織中TCTP表達(dá)情況,HE染色法觀察肝臟組織病理改變。 結(jié)果:1、與正常對(duì)照組相比,鮑曼不動(dòng)桿菌膿毒癥各組中性粒細(xì)胞和肝臟組織中TCTP表達(dá)量增高,且于12小時(shí)達(dá)高峰(P0.05);與鮑曼不動(dòng)桿菌膿毒癥各組相比,血必凈治療各組中性粒細(xì)胞和肝臟組織中TCTP表達(dá)量分別較對(duì)應(yīng)組降低(P0.05);2、與正常對(duì)照組相比,鮑曼不動(dòng)桿菌膿毒癥各組血漿中IL-6、IL-10含量增加,且于12小時(shí)達(dá)高峰(P0.05);與鮑曼不動(dòng)桿菌膿毒癥各組相比,血必凈治療各組血漿中IL-6表達(dá)量分別較對(duì)應(yīng)組降低(P0.05),而IL-10升高(P0.05);3、正常對(duì)照組大鼠的肝臟組織中,僅可見散在的TCTP陽性細(xì)胞且顏色較淺,而鮑曼不動(dòng)桿菌膿毒癥各組肝臟組織中可見大量的TCTP陽性細(xì)胞存在且顏色較深,血必凈治療組肝臟組織情況介于二者之間(P0.05);4、病理切片HE染色可見鮑曼不動(dòng)桿菌膿毒癥各組肝臟組織炎癥反應(yīng)、細(xì)胞損傷較明顯,血必凈治療各組則較其減輕,正常對(duì)照組未見明顯異常。 結(jié)論:1、TCTP在鮑曼不動(dòng)桿菌膿毒癥各組大鼠中性粒細(xì)胞及肝臟組織中的表達(dá)增加,且于12小時(shí)達(dá)高峰,TCTP可能在鮑曼不動(dòng)桿菌膿毒癥發(fā)生、發(fā)展過程中具有重要意義;2、血必凈注射液可減輕鮑曼不動(dòng)桿菌膿毒癥大鼠的炎癥反應(yīng),減少血漿中促炎性細(xì)胞因子IL-6的釋放,增加抑炎性細(xì)胞因子IL-10的釋放,并降低中性粒細(xì)胞及肝臟中TCTP的表達(dá)含量;3、血必凈注射液可能通過減少TCTP的表達(dá),繼而影響促炎性細(xì)胞因子IL-6的釋放而發(fā)揮抗炎作用。
[Abstract]:Objective: to study the expression of translation-controlled tumor protein (Thetranslationallycontrolledtumorprotein,TCTP) in neutrophils and liver tissues of Acinetobacter baumannii sepsis rats at different time points, and to detect the inflammatory factor IL-6, after intervention with Xiebijing. To investigate the relationship between TCTP and Acinetobacter baumannii sepsis and the effect of Xue Bijing on the expression of IL-10 and liver histopathology. Methods: 1Forty-two clean grade adult male wistar rats were randomly divided into 7 groups: the normal control group, (N), the control group was treated with acinetobacter baumannii sepsis for 6 h and 12 h, and the control group was treated with B6 h B12 h and B24 h, X6 h, X 12 h, X 24 h, and X6 h, X 12 h, X 24 h, respectively. Acinetobacter baumannii sepsis group and Xuebijing treatment group rats were intraperitoneally injected with Acinetobacter baumannii suspension and normal control group rats intraperitoneal injection of the same amount of normal saline; Xuebijing treated rats in each group were injected with Xuebijing via tail vein, and rats in other groups were injected with the same amount of normal saline. 2. Normal group, Acinetobacter baumannii sepsis group at different time points and Xuebijing treatment group, the liver was taken from carotid artery of rats. The expression of TCTP in neutrophil was detected by westernblot, the expression of inflammatory factor IL-6,IL-10 in plasma was detected by ELISA, the expression of TCTP in liver was detected by westernblot and immunohistochemistry, and the pathological changes of liver were observed by HE staining. Results: 1. Compared with the normal control group, the expression of TCTP in neutrophils and liver tissues of Acinetobacter baumannii sepsis groups increased, and reached the peak at 12 hours (P0.05). Compared with each group of Acinetobacter baumannii sepsis, the expression of TCTP in neutrophils and liver tissue of Xuebijing group was lower than that of corresponding group (P0.05). 2. Compared with the normal control group, the plasma IL-6,IL-10 content of Acinetobacter baumannii sepsis group increased and reached its peak at 12 hours (P0.05). Compared with each group of Acinetobacter baumannii sepsis, the expression of IL-6 in plasma of Xuebijing group was lower than that of corresponding group (P0.05), but IL-10 was increased (P0.05). 3. In normal control group, only scattered TCTP positive cells were found in the liver tissue and the color was lighter, while in Acinetobacter baumannii sepsis group, a large number of TCTP positive cells were found in the liver tissue and the color was darker than that in the control group. The liver tissue of Xuebijing treatment group was between the two groups (P0.05). 4. HE staining of pathological sections showed that the inflammatory reaction of liver tissue was obvious in each group of Acinetobacter baumannii sepsis, the cell damage was more obvious, the treatment group of Xue Bijing was lighter than that of the control group, and no obvious abnormality was found in the normal control group. Conclusion: 1 the expression of TCTP in the neutrophils and liver tissues of rats with Acinetobacter baumannii sepsis increased and peaked at 12 hours. TCTP may play an important role in the occurrence and development of Acinetobacter baumannii sepsis. 2. Xuebijing injection can reduce the inflammatory response of Acinetobacter baumannii sepsis rats, reduce the release of pro-inflammatory cytokine IL-6 in plasma, and increase the release of anti-inflammatory cytokine IL-10. The expression of TCTP in neutrophils and liver was also decreased. 3. Xuebijing injection may play an anti-inflammatory effect by reducing the expression of TCTP and then affecting the release of pro-inflammatory cytokine IL-6.
