IL-17、RORγt在百草枯中毒小鼠急性肺損傷中的表達(dá)

【摘要】:目的:觀察Th17型細(xì)胞因子IL-17A和特異性轉(zhuǎn)錄因子維甲酸相關(guān)孤獨(dú)受體(RORγt)在百草枯(PQ)中毒小鼠急性肺損傷的表達(dá)及意義。方法:雄性昆明小鼠隨機(jī)分為對(duì)照組和模型組,各8只。模型組予一次性腹腔注射20 mg/kg PQ建立染毒模型,對(duì)照組予同法注射等量生理鹽水,48 h后處死小鼠并取材。HE染色觀察肺組織病理變化;原位末端缺刻標(biāo)記法觀察細(xì)胞凋亡并計(jì)數(shù);酶聯(lián)免疫吸附法檢測(cè)血清IL-17A的水平;免疫熒光定量PCR法和蛋白質(zhì)免疫印跡法檢測(cè)肺組織RORγt m RNA及IL-17蛋白的表達(dá)。結(jié)果:與對(duì)照組相比,模型組小鼠肺組織炎癥及細(xì)胞凋亡顯著增加;血清IL-17A含量明顯增多(P0.05);肺組織RORγt m RNA和IL-17蛋白的表達(dá)均增高。結(jié)論:Th17可能通過(guò)分泌IL-17參與PQ中毒小鼠急性肺損傷的炎癥過(guò)程,并在其致病中發(fā)揮重要作用。
[Abstract]:Aim: to investigate the expression and significance of Th17 type cytokine IL-17A and specific transcription factor retinoic acid-associated lonely receptor (ROR 緯 t) in acute lung injury induced by paraquat (PQ) in mice. Methods: male Kunming mice were randomly divided into control group and model group with 8 mice each. The model group was injected intraperitoneally for 20 mg/kg PQ and the control group was injected with the same amount of normal saline. After 48 hours, the mice were killed and the pathological changes of lung tissue were observed by HE staining. Apoptosis and count were observed by in situ terminal dehiscence labeling, serum IL-17A levels were detected by enzyme-linked immunosorbent assay (Elisa), and the expression of ROR 緯 t m RNA and IL-17 protein in lung tissue were detected by immunofluorescence quantitative PCR and Western blot. Results: compared with the control group, the lung tissue inflammation and apoptosis in the model group were significantly increased, the serum IL-17A content was significantly increased (P0.05), and the expression of ROR 緯 t m RNA and IL-17 protein were increased in the lung tissue of the model group. Conclusion: Th17 may participate in the inflammatory process of acute lung injury in mice poisoned by PQ by secreting IL-17 and play an important role in its pathogenesis.
