ω-3多不飽和脂肪酸對燒傷大鼠腎臟及炎癥反應(yīng)的影響

【摘要】:目的:研究ω-3多不飽和脂肪酸對嚴(yán)重?zé)齻笫竽I臟及機(jī)體炎癥反應(yīng)的影響,,探討其對腎臟保護(hù)作用的可能機(jī)制。 方法:清潔級SD大鼠72只,雌雄不限,隨機(jī)分為3組:正常組(A組,n=8)、對照組(B組,n=32)和治療組(C組,n=32)。A組用溫水模擬燙傷過程,B組和C組采用大鼠30%TBSA Ⅲ度燒傷模型。A組大鼠經(jīng)尾靜脈注射生理鹽水2mL/(kg·d),B組每天定時(shí)經(jīng)尾靜脈注射20%力能中長鏈脂肪乳2mL/(kg·d),C組在相同時(shí)間點(diǎn)經(jīng)尾靜脈注射與B組等氮等熱量的尤文ω-3魚油脂肪乳2mL/(kg·d)。分別觀察傷前(A組)、傷后(B組、C組)1d、4d、7d、10d血清前列腺素2(PGE2)、內(nèi)皮素-1(ET-1)、白細(xì)胞介素-1(IL-1)和腫瘤壞死因子-α(TNF-α)的水平;RT-PCR檢測腎臟組織TNF-αmRNA的表達(dá);透射電鏡觀察腎組織形態(tài)學(xué)改變。每個(gè)時(shí)相點(diǎn)8只大鼠。 結(jié)果:1.B、C兩組在傷后各時(shí)相點(diǎn)血清PGE2、ET-1、IL-1、TNF-α水平顯著高于傷前(P0.01)。C組血清PGE2、ET-1、IL-1、TNF-α水平較B組均降低,在傷后4d、7d、10d與B組比較差異有顯著性(P0.05或P0.01)。2.透射電鏡觀察:與A組比較,B組傷后可見腎組織細(xì)胞核膜腫脹,線粒體腫脹且有空泡變,嵴突破壞;C組傷后可見腎組織細(xì)胞核膜腫脹(較B組減輕)或完整,線粒體腫脹(較B組減輕),嵴突破壞(較B組減輕)或完整。3. B組、C組在傷后各時(shí)相點(diǎn),腎組織TNF-αmRNA表達(dá)較A組明顯升高;與B組比較,在傷后4d、7d,C組腎組織TNF-αmRNA的表達(dá)明顯降低。 結(jié)論:1.ω-3PUFAs降低燒傷后血清PGE2、ET-1、IL-1、TNF-α的釋放。 2. ω-3PUFAs減輕燒傷后腎臟組織TNF-αmRNA的表達(dá)。 3.ω-3PUFAs可以減輕燒傷后腎臟組織的損傷。
