多臟器功能衰竭犬室性心律失常與KCNE1關(guān)系的研究

【摘要】:目的:探討多臟器功能衰竭(MODS)犬室性心律失常的發(fā)生與KCNE1基因的關(guān)系。方法:選擇健康雜種犬14只,雌雄不限。隨機(jī)分為MODS模型組7只,對(duì)照組7只。實(shí)驗(yàn)組失血性休克復(fù)蘇后靜脈注射內(nèi)毒素形成"二次打擊"制作犬MODS模型。當(dāng)MODS犬發(fā)生室性心律失常,搶救無(wú)效死亡后,迅速取出犬的心臟,剪下犬的左心室,置于液氨中固定,后轉(zhuǎn)移至-80℃冰箱中保存。提取總RNA,反轉(zhuǎn)錄為cDNA。采用實(shí)時(shí)熒光定量聚合酶反應(yīng)(qRT-PCR)法和蛋白免疫印跡(Western blot)方法檢測(cè)左心室KCNE1基因在mRNA和蛋白質(zhì)水平的表達(dá),研究MODS犬室性心律失常與KCNE1的關(guān)系。結(jié)果:與對(duì)照組相比,MODS模型組KCNE1的mRNA和蛋白質(zhì)表達(dá)水平顯著降低(P0.05)。結(jié)論:KCNE1與室性心律失常的發(fā)生具有相關(guān)性,其低水平表達(dá)可能是引起MODS犬發(fā)生室性心律失常的重要因素。
