抗氧化劑對(duì)熱打擊人臍動(dòng)脈血管平滑肌細(xì)胞收縮反應(yīng)的影響

【摘要】:目的研究抗氧化劑對(duì)熱打擊過(guò)程中人臍動(dòng)脈血管平滑肌細(xì)胞(HUASMCs)收縮反應(yīng)的影響。方法將HUASMCs分為對(duì)照組、熱打擊組(HS組)和熱打擊前超氧化物歧化酶(SOD)預(yù)處理組(SOD+HS組),每組分別用低濃度NE(0.05mg/L)及常規(guī)濃度NE(1.0mg/L)刺激。各組細(xì)胞于41℃恒溫水箱中分別培養(yǎng)0.5、1、1.5、2h后添加NE刺激,采用細(xì)胞表面積收縮比(%)反映細(xì)胞對(duì)NE的反應(yīng)性。結(jié)果與對(duì)照組比較,無(wú)論是給予低濃度或常規(guī)濃度NE刺激,HS組細(xì)胞熱打擊1h時(shí)對(duì)于NE的反應(yīng)性均明顯增高,2h時(shí)對(duì)于NE的反應(yīng)性則明顯降低(P0.05或P0.01);SOD+HS組細(xì)胞對(duì)NE的反應(yīng)性在熱打擊至2h后均出現(xiàn)明顯下降(P0.05或P0.01)。對(duì)照組及HS組細(xì)胞對(duì)不同濃度NE刺激的反應(yīng)性差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),SOD+HS組細(xì)胞在熱打擊過(guò)程中對(duì)常規(guī)濃度NE的反應(yīng)性明顯高于對(duì)低劑量NE的反應(yīng)性(P0.05或P0.01)。無(wú)論給予常規(guī)濃度或低濃度NE刺激,SOD+HS組細(xì)胞對(duì)NE的反應(yīng)性均低于HS組(P0.05或P0.01)。結(jié)論熱打擊過(guò)程中,HUASMCs對(duì)NE的反應(yīng)性呈時(shí)相性變化?寡趸瘎┰谥惺钤缙诳赡芸梢约m正HUASMCs對(duì)NE的過(guò)反應(yīng)現(xiàn)象,但在熱打擊中后期,抗氧化劑并不能改善其反應(yīng)性的下降。
